摘要
目的建立以β-环糊精(β-cyclodextrin,β-CD)和离子液体1-丁基-3-甲基咪唑-L-乳酸盐(1-bu-tyl-3-methylimidazoliumL(+)lactic acid,BMIM-LA)联合作为手性选择剂,用于分离扑尔敏、卡维地洛和文拉法辛对映体的毛细管电泳方法。方法采用未涂层熔融石英毛细管柱,背景电解质为Tris-HCl缓冲溶液,运行电压20 kV。考察了手性选择剂β-CD和离子液体的浓度、pH值、缓冲溶液浓度对对映体分离的影响。结果在最优化的电泳条件下,扑尔敏可达到完全分离,卡维地洛和文拉法辛部分分离,分离度分别为2.1、1.2和0.75。结论β-CD与离子液体联用可提高扑尔敏、卡维地洛和文拉法辛对映体的分离选择性。
Objective To separate the enantiomers of chlorphenamine,carvedilol and venlafaxine by capillary electrophoresis using β-cyclodextrin(β-CD)and chiral ionic liquid 1-butyl-3-methylimidazolium L(+)lactic acid(BMIM-LA)as chiral selectors.Methods The separations of enantiomers were performed in an untreated fused-silica capillary tube,in the buffer of Tris-HCl at room temperature,a high voltage of 20 kV was applide.The effect of the concentrations of BMIM-LA and β-CD,buffer pH and the concentration of the buffer on the separation were investigated.Results Under the optimal conditions,the resolutions of chlorphenamine,carvedilol and venlafaxine enantiomers were 2.1,1.2 and 0.75,respectively.Conclusions The synergistic interaction of β-CD and BMIM-LA improves the separation effect and provides a new method for the separation of chiral drugs.
