摘要
目的:观察高压氧联合放射性药物^(32)P胶体对大鼠移植瘤的治疗作用。方法:完成实验动物肿瘤模型的制作后,将实验动物随机分为对照组、高压氧治疗组、^(32)P胶体治疗组、^(32)P胶体和高压氧治疗组(联合治疗组),每组10只。治疗后观察各组肿瘤生长情况,测定肿瘤第7天、12天、20天时的体积,并计算其生长率;通过病理观察各组肿瘤组织光镜下形态特征;通过免疫组化技术检测肿瘤组织的Caspase-3的阳性表达率、对CD_(34)阳性血管进行微血管密度(MVD)计数。结果:对照组、高压氧治疗组、^(32)P胶体治疗组、联合治疗组第12天、20天的肿瘤生长率分别为(223.21±68.43)%、(899.45±439.63)%;(219.32±80.87)%、(889.23±625.14)%;(163.56±58.73)%、(505.07±342.69)%;(103.81±41.57)%、(114.68±41.83)%,联合治疗组明显小于其它各组(P<0.05)。联合治疗组的MVD计数明显小于其它各组(P<0.05)。联合治疗组的Caspase-3的阳性表达率明显高于其它各组(P<0.01)。结论:高压氧联合放射性药物^(32)P胶体可显著抑制大鼠移植瘤的生长,肿瘤血管新生,上调Caspase-3的表达,从而抑制癌细胞增殖、促进细胞凋亡。
Objective: To observe the effect of combination of hyperbaric oxygen with 32 P-colloid on rats with implanted tumor. Methods: Walker-256 cancerous ascites injection was used to make a rat tumor-bearing model. Then the rats were randomly divived into control group, hyperbaric oxygen group,32 P-colloid group, hyperbaric oxygen and 32 P-colloid group (combination therapy group) with 10 rats in each group. The tumor growth, tumor volume on 7th,12th and 20th day were observed to calculate its growth rate after treatment. The morphological changes were observed by pathological examination. The microvessel density(MVD) and Caspase-3 were detected by immunohistochemistry method. Results: The tumor growth rates on the 12th and 20th day in control group, hyperbaric oxygen group ,32 P-colloid group and combination therapy group were (223.21 ± 68.43) %, ( 899.45 ± 439.63 ) % ; (219.32 ± 80.87) % ,889.23 ± 625. 14)% ;(163.56 ±58.73)% ,(505.07 ±342.69)% ;(103.81 ±41.57)%,(114.68 ±41.83)% respectively. The tumor growth rate of combination therapy group was significantly lower than those of the other three groups(P 〈 0. 05). The MVD of combination therapy group was significantly less than those of the other groups (P 〈 0. 05 ). The positive expression rate of Caspase-3 in combination therapy group was obviously higher than that of the other groups(P 〈 0.01 ). Conclusion: Hyperbaric oxygen combined with radioactive 32p-colloid can inhibite the growth of tumor xenograft of rats and the angiogenesis of blood vessels as well as increase the expression of Caspase-3 signifi- cantly. It can thus inhibit the proliferation and promote the apoptosis of cancer cells.
