摘要
目的:探讨ICAM1 相关肽抑制角朊细胞将超抗原TSST1 递呈给T淋巴细胞。方法:应用鼠抗人CD54 单克隆抗体筛选噬菌体随机十五肽库,经4 轮筛选后,挑取20 个克隆进行测序,利用含保守序列的阳性克隆噬菌体对角朊细胞递呈超抗原TSST1 给T细胞进行阻断试验。结果:得到保守序列为ITRSFQSSMEPGPPG,所筛选的阳性噬菌体克隆与野生型噬菌体相比具有明显的阻断效应( P< 001) 。结论:ICAM1 相关小分子肽可望降低超抗原介导的皮肤炎症反应。
Objective:To explore the inhibiting effect of ICAM 1 related sequences peptide on keratinocytes which present superantigen TSST 1 to T cells.Methods:Used mouse anti human CD54 McAb to screen phage display 15 mer peptide random library.After 4 round selections chose 20 clones at random and analyzed the DNA sequences.Used the phage contained conservative sequences to block that keratinocytes present superantigen TSST 1 to T cells.Results:The conservative amino acid sequence is ITRSFQSSMEPGPPG.The blocking effect of positive colonal phage is significantly stronger compared with that of wild type phage(P<0 01).Conclusion:ICAM 1 related small peptide could reduce skin inflammation mediated by superantigens.
