电针后大鼠中脑腹侧部粗提液对体外培养的多巴胺能神经元生长发育的影响 被引量:7

Effects of the Crude Extracts Prepared from the Ventral Mesencephalic Region of Rats Stimulated with Electro-acupuncture on the Growth and Differentiation of Dopaminergic Neurons in Vitro

摘要 电针正常大鼠后摘取其中脑腹侧部,制备电针后大鼠中脑腹侧部粗提液,加入到体外原代培养的中脑腹侧部神经元中。分别在第5天和第10天用免疫细胞化学方法显示多巴胺能神经元,观察电针后大鼠中脑腹侧部粗提液对体外培养多巴胺能神经元表达的数目、神经元胞体直径和神经突起长度的影响。证实电针后大鼠中脑腹侧部粗提液能够促进多巴胺能神经元胞体的发育和突起的生长,但对多巴胺能神经元表达的数目无明显影响。提示这种作用的产生可能与电针刺激了中脑腹侧部某些具有神经营养作用的物质的产生或分泌有关。 Normal SD rats were stimulated for a period of time with electro-acupuncture and then were killed. The ventral mesencephalic regions of the rats were excavated to prepare the crude extracts, which, afterwards, were added into the primary mesencephalic neurons in culture. An immunocytochemical staining was performed to detect the dopaminergic neuron on the 5th and the 10th day respectively in vitro, and the average expression number of the positive neurons in each well was counted, the diameter of the cell body and the length of the neurite were measured in each group. The results indicated that the crude extracts made from the ventral mesencephalic regions of rats stimulated with electro-acupuncture could promote the development of cell body and the growth of the neurite but had no effect on the expression number of dopaminergic neurons. It is suggested that the results might be probably related to the effect of electro-acupuncture in stimulating the release or secretion of some potential neurotrophic factors in the ventral mesencephalic region.
