摘要
2009年12月从疑似新城疫的商品肉鸡中分离到一株具有血凝性的病毒,经血清学和RT-PCR鉴定,确认为鸡新城疫病毒。该纯化病毒的MDT(最小致死量引起所有鸡胚死亡的平均时间)为56 h,ICPI(1日龄SPF鸡脑内接种分离病毒致病指数)为1.76,ELD50(鸡胚半数致死量)为10-8.5;序列分析表明,该分离株F蛋白基因长1662 bp,编码553个氨基酸,推导的F基因氨基酸序列裂解位点为112R-R-Q-K-R-F117,具备强毒的典型特征。经对F基因的全序列分析,该病毒与鸡新城疫病毒F48E9株,La Sota株的核苷酸同源性分别为85.9%、83.4%,与TCQQ/Tianjin/08株、CJG/xinjiang/07株核苷酸同源性为99.6%,具备基因Ⅶ型新城疫病毒的分子特征,命名该毒为APMVl/chicken/China/HN68/09。
A virus which could agglutinate chicken red blood cell from broiler was isolated in December,2010.It is identified as Newcastle Disease virus by serology and RT-PCT experiment.The purified virus'ELD50(the Embryo median lethal dose),MDT(Mean death time of embryos),ICPI(intracerebral pathogenicity index in 1-day-old chickens) was 10-8.5/0.1 mL,56 h,1.76,respectively.The sequencing analysis showed that the F gene encoding 553 amino acids was 1662 bp in length.The deduced amino acid sequences of F gene cleavage site was 112R-R-Q-K-R-F117,which was with the typical characteristic of virulent.Compared with F48E9,La Sota NDV strains,there were 85.9 % and 83.4 % of homogeneity,respectively,and was closer with the virus strains from Tianjin and Xinjiang.The isolation was with genotype VII molecular characterization of Newcastle disease virus,was named as APMVl /chicken/China/HN68/09.
