摘要
利用去甲肾上腺素对牛血红蛋白模拟酶催化体系的抑制作用,建立了酶催化动力学光度法测定去甲肾上腺素的新方法.研究了以酸性铬蓝K作为氢供体底物的酶催化反应体系的催化特性和反应条件,其抑制程度与去甲肾上腺素浓度成线性关系,测定的线性范围为7.2×10^(-8)~7.2×10^(-6)mol/L,检出限为2.4×10^(-9) mol/L.对浓度为6.0×10^(-6) mol/L的去甲肾上腺素进行11次平行测定,其相对标准偏差为4.1%.该方法已成功的应用于制剂中去甲肾上腺素含量的测定,结果满意.
A high sensitive and simple spectrophotometrie method for the determination of noradrenaline based on the inhibition of hemoglobin for the reaction of H202 and acid chrome blue K was developed. The percentage inhibition (I%) of system is calculated under the optimal experimental conditions. The calibration curve is linear in the range of 7.2× 10^-8- 7.2 × 10^-6mol/L with the detection limit of 2.4×10^-9mol/L. The relative standard deviation of this method is 4.1% at 6.0 × 10-6 mol/L for 11 determinations. This method can be used for the determination of noradrenaline in pharmaceuticals with satisfactory results.
