不同基因型矮牵牛高频再生体系建立的研究 被引量:4

摘要 为建立不同矮牵牛品种的高频再生体系,以10个矮牵牛品种为材料,研究了不同基因型、不同生长调节剂配比、不同接种方式、叶片不同部位和不同外植体对不定芽再生的影响。研究结果表明:基因型是影响不定芽再生的重要因素之一,从10个品种中筛选获得了5个品种的高频再生体系;6-BA和NAA及其配比极显著影响矮牵牛不定芽再生率和平均不定芽再生数量,其中0.2 mg.L-1NAA极显著的促进了矮牵牛不定芽再生,其次是0.1 mg.L-1NAA,而高浓度NAA抑制不定芽再生;不同品种适宜的分化培养基不同,筛选获得了不同品种适宜的分化培养基;不同接种方式对不同品种不定芽再生影响不同,其中不同接种方式对黄金紫罗兰的不定芽再生有极显著影响,对黄金肉红、珊瑚红则没有影响;叶片不同部位极显著影响黄金紫罗兰星条不定芽的再生,对其它品种没有影响;叶片外植体是矮牵牛不定芽再生的最佳外植体。 The effects of different genotypes,hormone conditions,inoculation modes,parts of leaves and explants on shoot regeneration of ten Petunia hybrida cultivars were studied in order to eatablish high frequence regeneration system.The results indicated that genotype was an important factor for shoot regeneration.In this study,the high frequence regeneration of five cultivars were obtained from ten cultivars;And shoot regeneration and the average regeneration number were influenced highly by 6-BA、NAA and different hormone conditions.Shoot regeneration were promoted highly on culture medium including 0.2 mg·L-1NAA,and then on 0.1 mg·L-1NAA;Different culture media were suited for different cultivars;Inoculation mode made significant influences on shoot regeneration of cultivars 'Huangjinziluolanxingtiao',but no effects on 'Huangjinrouhong'and'Shanhuhong';shoot regeneration of 'Huangjinziluolanxingtiao' were also influenced significantly by different part of leaves and explants,which did not make effects on other cultivars;Leaves were the best explants suited for shoot regeneration of Petunia hybrida.
