摘要
目的 探讨听神经瘤中神经生长因子受体的表达与肿瘤自然生长率、细胞增殖和血管形成的关系。方法 运用免疫组化技术,检测34 例人听神经瘤中神经生长因子受体蛋白、增殖细胞核抗原和微血管密度的表达。分析神经生长因子受体与增殖细胞核抗原、微血管密度及听神经瘤生长率的关系。结果 神经生长因子受体在所有肿瘤组织中表达(100 % ) ,而在正常听神经组织中无表达。肿瘤明显生长组神经生长因子受体、增殖细胞核抗原表达水平和微血管密度平均值显著高于缓慢或无生长组( P< 005 , P< 005 , P< 001) 。神经生长因子受体且和微血管密度( P< 001) 密切相关,但与增殖细胞核抗原( P> 005) 无关。结论 神经生长因子受体在明显生长的听神经瘤中表达增强,且与肿瘤的血管生成密切相关,提示神经生长因子受体参与了听神经瘤的发生过程。
Objective To investigate the relationship of nerve growth factor receptor (NGFR) with tumor proliferation and angiogenesis in human acoustic neuroma(AN).Methods NGFR, proliferation cell nuclear antigen (PCNA) expression and microvessel density(MVD) visualized by the endothelial marker UEA in 34 cases of human AN were examined by using immunohistochemical staining.NGFR, PCNA and MVD expressions were analysed together with their relation to annual growth index of AN.Results Immunoreactivity for the NGFR was observed in all cases (100%).No expression of NGFR was observed in normal acoustic nerves. The positive immunoreactivity for NGFR,number of PCNA positive nuclei and MVD were significantly higher in macroscopic growing tumors than in slow or zero growing tumors (P<0 05,P<0 05,P<0 01).NGFR was statistically related to MVD(P<0 01).No significant relation was observed between NGFR and the proliferation, as estimated by PCNA (P>0 05).Conclusion The macroscopic growth of AN was associated with strong positive expression of NGFR which was related to angiogenesis of AN.The results indicate that NGFR may be involved in the pathogenesis of AN.
