摘要
目的筛选高良姜素的最佳醇提和初步纯化工艺条件。方法以高良姜素含量为指标,采用高效液相色谱法,通过L9(34)正交试验设计优选醇提工艺条件,确定高良姜的最佳醇提工艺;以高良姜素的纯度为指标,采用不同酸碱水溶液对醇提液进行初步纯化。结果高良姜素的最佳醇提工艺条件为乙醇浓度80%,提取次数2次,溶剂倍数8倍,提取时间1.5 h;纯化工艺为3倍量的水溶液,静止24 h。结论优化的高良姜醇提工艺条件合理可靠,重复性良好;纯化工艺条件操作方便,除杂效果好,为后续纯化实验提供便利。
Objective To explore the optimum method for the extraction and purification of Galangin from Alpinia officinarum Hance. Methods Using the content of Galangin which was determined by HPLC as index, the optimum ethanol extraction of Alpinia officinarum Hance was studied with orthogonal test of L9(3^4). With the puirty of Galangin as index, the fluid was purified with different acidic and alkaline water. Results The optimum extracting conditions was confirmed, they were as 8 times amount of 80% ethonal, two times and 1.5 h per time. The optimum purification were as adding 3 times amount of water, standing for 24 h. Conclusion The optimum technique for the extraction and purification was rational and feasible, which can help the follow-up study.
