尿毒清胶囊含药血清对人近端肾小管上皮细胞结缔组织生长因子表达的影响 被引量:5

摘要 目的观察正常人的尿毒清胶囊含药血清对尿毒症血清刺激的人肾小管细胞(HK-2)表达结缔组织生长因子(CTGF)的影响。方法将体外培养HK-2细胞分为6组:正常对照组、5%尿毒症血清组、10%尿毒症血清组、5%尿毒清胶囊含药血清组、10%尿毒清胶囊含药血清组和蒙诺组,用逆转录-聚合酶链反应(RT-PCR)测定CTGFmRNA的表达量。结果各尿毒症血清组、含药血清组和蒙诺组CTGF mRNA的表达较正常对照组均有极显著性升高(P<0.01),其中10%尿毒血清组最高,与其它各组比较均有极显著差异(P<0.01),5%含药血清组与5%尿毒症血清组无差异(P>0.05),而10%含药血清组和蒙诺组较5%尿毒症血清组均有显著性下降(P<0.05),5%和10%含药血清组与蒙诺组之间比较差异均无显著性(P>0.05)。结论尿毒清胶囊抗肾纤维化作用的可能机制是抑制参与肾纤维化的肾小管细胞强致纤维化因子的产生。 Objective To study the effect of Niaoduqing serum on expression of connective tissue growth factor in HK-2 cells.Methods Human renal tubular epithelial cells(HK-2) were respectively cultured in six different media and were diveded into normal control group,5% uremic serum group,10% uremic serum group,5% NDQC serum group,10% NDQC serum group and Monopril group(Monopril concentration was 10-6mmol/L).Connective tissue growth factor messenger RNA(CTGFmRNA)was detected by reverse transcription-polymerase chain reaction(RT-PCR) method.Results The expression of CTGFmRNA was much higher in uremic serum groups,NDQC serum groups and Monopril group than in normal group(P0.01).The expression in 10% uremic serum group was highest in all groups(P0.01).No significant difference was found in 5% NDQC serum group compared with that in 5% uremic serum group(P0.05).CTGFmRNA expression was remarkably lower in 10% NDQC serum group and monopril group than in 5% uremic serum group(P0.05).No significant difference of CTGFmRNA expression was found among 5% NDQC serum group,10% NDQC serum group and monopril group(P0.05).Conclusion The mechanism of NDQC against renal fibrosis possibly relates to preventing cells from synthetizing or secreting CTGF.This research result will provide experimental evidence for NDQC ' s clinical application.
