摘要
目的建立测定陈皮中橙皮苷含量的微波辅助萃取结合高效液相色谱的方法。方法通过对微波辅助萃取提取条件进行优化,确定了萃取的最佳条件:提取溶剂为50%二甲亚砜(DMSO)水溶液,固液比1:10,提取功率400 W,提取时间3 min。采用C18色谱柱,流动相为甲醇-0.2%三氟乙酸溶液(69:31,V/V),检测波长为283 nm。结果橙皮苷在0.7-14.0μg.mL-1浓度范围内线性关系良好(r=0.999 7)。平均回收率为101.75%,RSD为2.53%。结论该方法简便、快速、准确,适用于分析测定陈皮中橙皮苷的含量。
Objective A method of microwave assisted extraction followed by HPLC was established for the determination of hesperidin of pericarpium citri reticulatae. Methods Through optimization of microwave assisted extraction method,the optimum conditions were as follows: extraction solvent of 50% DMSO aqueous solution,solid-liquid ratio of 1:10,microwave power of 400 W,irradiation time of 3 min.A C18 column was used with the mobile phase of methanol-0.2% trifluoroacetic acid solution(69:31,V/V) at the detection wavelength of 283 nm. Results The calibration curve was linear in the range of 0.7-14.0 μg·mL-1.The average recovery was 101.75% with RSD of 2.53%. Conclusion This method was simple,fast,accurate,and it is adapted to determine hesperidin of pericarpium citri reticulatae.
