摘要 分别用乙醇、乙酸乙酯、丙酮提取姜黄粉中的姜黄素,通过正交实验确定最佳提取溶剂为丙酮.采取逆向思维,将活性白土用于姜黄色素脱油脂进行提纯,并建立了高效液相色谱外标法用于分析姜黄素的含量.实验表明:用丙酮提取的,提取率最高,可达94.57%.丙酮提取液流经活性白土层析柱后,测得活性白土对姜黄素的吸附容量为7.34%,分别采用碱性水、碱性乙醇、碱性丙酮洗脱被吸附的姜黄素.通过单因素实验确定最佳吸附和洗脱条件.最终可得到高纯度、不吸潮的精制姜黄素产品.实验结果显示,色素的产率为2.36%,产品纯度为94.29%,色素总收率达80.71%. The curcumin was respectively extracted from curcuma powder by alcohol,ethyl acetate and acetone.It was determined that acetone was optimal extracting solvent through the orthogonal tests.This study adopted reverse thinking,using activated clay for pigment-degreasing to purify curcumin,and established HPLC external standard method for the determination of curcumin content.The experiment indicated that the extraction rate was the highest with acetone extraction,up 94.57%.The absorption capacity of activated clay for curcumin was 7.34% when the extract flowed through the activated clay chromatographic column.Three alkalized solvents,water,ethanol and acetone,were respectively used to elute the absorbed curcumin from the column.The optimal adsorption and elution condition was determined by single factor experiments.The non-hygroscopic refined curcumin product with high-purity could be obtained eventually.The experiment results showed that the product rate of curcumin was 2.36 %,product purity 94.29%,and total pigment recovery rate 80.71%.
