摘要
比较了食蟹猴2型糖尿病(T2DM)不同发病阶段与正常对照组的空腹血糖值(FPG)、甘油三酯(TG)、胆固醇(CHOL),低密度脂蛋白胆固醇(LDL-C),高密度脂蛋白胆固醇(HDL-C)在血液中含量,及荧光定量PCR方法测定的NF-κB和IκBα在外周血白细胞中表达量。结果显示,FPG、CHOL和LDL-C随疾病进行呈上升趋势,而HDL-C先上升后下降,血糖血脂值变化表明,在T2DM临床前期就出现脂代谢异常;除IκBα表达量在模型初期显著降低,NF-κB表达量在临床期显著升高外,其他各阶段无差异,基因表达量变化表明,在疾病初期开始启动IκB/NF-κB信号通路,到临床期启动完全。根据以上结果推测,前期低水平炎症反应引起脂代谢异常,而后者又促进前者的发展,至临床期才出现明显炎症状态。
The contents of FPG,TG,CHOL,LDL-C and HDL-C in peripheral blood was measured,and the expression of IκBα and NF-κB in peripheral blood leucocytes was examined by using quantitative PCR in different phases of Cynomolgus Type 2 diabetes mellitus(T2DM) and normal control group.The results indicated that the contents of FPG,CHOL,LDL-C increased with the progression of T2DM,while HDL-C increased in early period then declined after the preclinical period,all the changes demonstrated that abnormal glucolipids metabolism began to appear in the early period of the disease.The expression level of IκBα reduced significantly in initial model phase,and the expression level of NF-κB increased significantly in clinical phase,these changes showed that NF-κB and IκBα played an active role in the process of T2DM and the Iκ Bα/NF-κB signaling pathway was activated from initial model phase to clinical phase.Based on the above results,we speculated that the low-grade inflammatory response in early period might cause dyslipidemia,which in turn promote the development of the former,until in the clinical phase appeared the typical inflammatory response.
