细菌体内诱生抗原筛选技术的研究进展

摘要 只在进入宿主体内后表达而在体外不表达的基因称为体内诱生基因。大量研究表明,这些基因对病原菌进入体内的生存和致病具有重要的意义。体内诱生基因的筛选可为抗菌药物的开发、临床诊断试剂的研究、疫苗的设计提供新的准确靶标,从而为预防和治疗工作奠定基础。近年来有关病原菌体内诱导基因的研究越来越受到重视,人们发展了多种体内诱生基因的筛选技术,如体内表达技术(IVET)、信号标签突变(STM)、体外转座进行基因组分析和作图(GAM-BIT),这些方法因需建立动物模型而使其使用受限,体内诱生抗原技术(IVIAT)则因具有不依赖于动物模型、简便、快捷、高通量而得到了广泛的应用。 Those genes that express only in the host cells are frequently referred to as in vivo induced antigen.It has been demonstrated in previous studies that these genes play a significant role in the survival and pathopoiesis of the pathogen in the host.The screening of in vivo induced antigen could supply new accurate target in the development of antibacterials,the research of clinical diagnostic reagent,the contrive of vaccine,accordingly lay a solid foundation on prevention and therapy.These years,increasing attention has been paid to the study of the in vivo induced antigen,and numerous screening techniques has been established: in vivo expression technology(IVET),signature-tagged mutagenesis(STM),genomic analysis and mapping by in vitro transposition(GAM-BIT),but these techniques are limited due to the animal model.However,in vivo induced antigen technology(IVIAT) has been widely applied,for it is independent on the animal mode,simple,swift and high-flux.
