抗P37多克隆抗体的制备 纯化与鉴定

Preparation and Identif Ication of Polyclonalantibody Against P37

目的:用P37融合蛋白免疫动物制备抗血清,为猪鼻支原体感染作用的分子机制提供依据。方法:诱导表达GST-P37蛋白并以其免疫兔制备免疫血清,ELISA法检测抗血清的效价;Western blot-ting检测抗血清特异性。结果:免疫家兔并纯化抗血清得到特异的抗P37抗体,该抗体能检测到P37蛋白的表达。结论:成功制备了抗P37的多克隆抗体,对进一步研究P37的功能提供了一个有用的工具。

Objective：To offere some prophase works by preparing polyclonal antibody,in order to study molecular mechanism of P37.Methods：The recombinant plasmid was transformed into E.coli BL21 to express the fusion p rotein of GST-P37.The polyclonal anti-P37 antibody were prepared by immunizing rabbits with the purified protein.The preimmune serum was as the negative control during the ELISA assay of antiserum valency.The specificity of the antiserum was identified by Western blotting.Results：The polyclonal antibody showed specific response to P37 by Western blot analysis.These results suggested that the polyclonal antibody can be used for further studies of P37 function.Conclusion：A polyclonal anti-P37 antibody has been synthesized successfully.
