摘要
目的探讨不同低氧环境对大鼠肺动脉平滑肌细胞(pulmonary arterial smooth musclecells,PASMCs)增殖活性的影响。方法将原代培养的大鼠PASMCs分为常氧组(21%O2,5%CO2)、低氧组(10%O2,85%N2,5%CO2)、缺氧组(1%O2,94%N2,5%CO2)进行培养。通过细胞计数和四甲基偶氮唑蓝(MTT)自动化比色法测定以上各组PASMCs的增殖活性。结果与常氧组和低氧组相比,缺氧组2天后PASMCs明显增殖(P<0.01),5天后细胞数达到顶峰,6天后缺氧组PASMCs开始出现空泡化和死亡,细胞数开始减少。低氧组PASMCs的生长基本和常氧组一样,两者相比无显著性差异。结论离体培养的大鼠PASMCs只有在比较低的氧环境(1%O2,94%N2,5%CO2)下才能产生明显的增殖作用。
Objective To explore the effect on different hypoxic proliferation of rat pulmonary arterial smooth muscle cells(PASMCs).Methods The primary culture PASMCs of rat were randomly divided into three groups,including: normoxia group(21 % O2,5 % CO2),hypoxia group(10 % O2,85 % N2,5 % CO2)and anoxia group(1 % O2,94 % N2,5 % CO2).The proliferation was tested by cell count and methyl thiazolyl tetrazolium(MTT) method.Results The PASMCs count of anoxia group was more than normoxia group and hypoxia group after 2 days,and the proliferation of PASMCs were more obvious in anoxia group than normoxia group and hypoxia group after 5 days(P 〈 0.01).The PASMCs in anoxia group occurred vacuolization and apoptosis from 6 days.Conclusion There is more lower hypoxia which may stimulate PASMCs and promote proliferation.
