摘要
综合利用多种生物信息学工具对罗伊氏乳杆菌亚油酸异构酶基因及其启动子区进行了定位,分析了4种乳酸菌亚油酸异构酶基因的上游调控区,发现了主要保守位点.分析证明罗伊氏乳杆菌亚油酸异构酶具有2个二硫键,1个糖基化位点,多个磷酸化位点,无信号肽序列,但在25(26)-42位存在跨膜区.证明罗伊氏乳杆菌亚油酸异构酶的密码子偏好性与大肠杆菌和酿酒酵母的密码子偏好性存在差异,初步分析了该基因在这些宿主菌中表达时可能存在的问题,并提出了改造方案.
We comprehensively utilized many kinds of bioinformatics tools to locate the linoleic acid isomerase gene and it′s promoter region,analyzed the upstream regulatory region of linoleic acid isomerase genes of four kinds of Lactobacillus,and found the main conservative sites.The analysis testified that the linoleic acid isomerase of Lactobacillus reuteri had two disulfide bonds,one glycosylation site,multiple phosphorylation sites,no signal peptide sequence,and had transmembrane domain at 25(26)-42 site.The results showed that the codon bias of the Lactobacillus reuteri was different from that of Escherichia coli and Saccharomyces cerevisiae.In addition,we primarily analyzed the problem might exist during expression of the gene in the host strain,and proposed the improvement plan.
