补肾活络方对PCOS大鼠促排卵及卵巢IGFBP-1表达影响的研究

Study on the Mechanism of BushenHuoLuofang in Ovulation promting to Rats with Polycystic ovary and insulin-like growth factor-binding proteins-1 expression in the Ovary

目的：探讨以补肾活络立法的＂补肾活络方＂对肾虚血瘀型不孕症促进卵泡发育及促排卵的可能机制。方法：建立PCOS大鼠不孕模型,运用分子生物学和显微形态学技术,对正常组、模型组、中药组、西药组SD雌大鼠卵巢细胞IGFBP-1基因表达水平进行检测,同时观察卵巢局部超微形态学的变化。结果：中药组、西药组与模型组在卵巢重量及卵泡数目上比较,差异有统计学意义（P〈0.05）;中药组与西药组在各级卵泡数目上比较,差异有统计学意义（P〈0.05）。模型组IGFBP-1mRNA的表达无论在表达面积和表达强度上都不及正常组、中药组和西药组（P〈0.05）;中药组与正常组在表达面积和表达强度上比较,差异无统计学意义（P〉0.05）。结论：以补肾活络立法的补肾活络方可以显著增加大鼠卵巢重量及卵泡总数;且通过影响大鼠卵巢局部IGFBP-1mRNA的表达,促进卵泡发育,从而达到促排卵的目的。

Obejective： To study the mechanism of BushenHuoLuofang based on the role of tonifying kidney.d issolving blood stasis treating female the sterility due to asthenia of the kidney deficiency and blood stasis and interpret the intervention role of promoting the growth of follicle ovulation.Methods：Pcos model was set up and screened by vaginal smear.The use of morphological techniques in molecular biology and microscopy,the normal group,model group,chinese medicines group,western medicine group SD female rats ovarian IGFBP-1 gene expression in cells were detected,while observing the morphological changes of ovary AMD.Results：The Chinese Medicines Board,WM group with the model group in ovarian weight and the number of follicles at all levels had a significant difference.（P0.05）Chinese medicine with western medicine group at all levels of follicle number had a significant difference（P0.05）.Model group LHRmRNA expression in the expression of both size and intensity of expression and not on the normal group,Chinese medicine and western medicine group（P0.05）.Chinese medicine group and the normal group in the area and the expression on the expression of no difference（P0.05）.Conclusion：BushenHuoLuofang based on the role of invigorating the kidney can greatly increase the weight of ASR rats＇ ovary and the quantities of ovarian follicles in ovary.By affecting the expression of the IGFBP-1mRNA in ovary,it improves the growth.
