摘要 目的观察Leptin受体(OB-R)在实验性脂肪肝与肝硬化大鼠中肝脏mRNA的表达情况,探讨OB-R在实验性脂肪肝与肝硬化发生和发展中的作用。方法 (1)建立实时荧光定量逆转录聚合酶链反应(FQ RT-PCR)技术检测OB-R的mRNA表达水平的方法及标准曲线;(2)采用FQ RT-PC检测非酒精性脂肪肝组、肝硬化组及对照组中OB-R的mRNA表达水平,从定量的角度研究OB-R及OB-R mRNA与非酒精性脂肪肝和肝硬化的关系以及它们二者的关系,以探讨表达与非酒精性脂肪肝、肝硬化的相关性及其在实验性脂肪肝与肝硬化发生和发展中的作用。结果非酒精性脂肪肝组、肝硬化组和对照组均有OB-R的mRNA表达,三者比较差异无统计学意义(P>0.05)。结论 OB-R的mRNA在非酒精性脂肪肝、肝硬化和对照组中均有表达,但其可能在脂肪肝和肝硬化的发生和发展中并没有起到直接作用,可能还有其他因子的共同参与。提示OB-R可能参与了肝脂肪变性形成,促进肝纤维化的发生。 Objective To observe the expression of the Leptin receptors gene in experimental fatty liver and cirrhosis liver,and to discuss the possible mechanisms of OB-R in experimental fatty liver and cirrhosis liver.Methods 1.We established a method of fluorescent quantitative reserve transcription polymerase chain reaction(FQ RT-PCR) to test the exppresion level of mRNA of Leptin receptors(OB-R) and its standard curve.2.The message RNA(mRNA) expression levels were measured by the FQ RT-PC method and compared quantitatively between non-alcoholic fatty liver(NAFL) model,cirrhosis liver model and the control group.Results The mRNA levels of Leptin receptors gene increased in the liver of experimental NAFL and cirrhosis liver.There was no significant difference between the mRNA levels of Leptin receptors gene in the NAFL group,cirrhosis liver group and control group(P〉0.05).Conclusion The mRNA levels of Leptin receptors gene were all expressed in NAFL and cirrhosis liver and the control group.This data do not support a role for leptin receptor in the pathogenesis of NAFL and cirrhosis liver.These observations proved Leptin receptors might appear to promote pro-fibrogenic responses in the liver.Our study results not only provide experimental basis for us to research the mechanisms of fatty liver and cirrhosis liver,but also establishe the theoretical foundation to prevent and treat all types of hepatic injury.
