摘要
目的:观察信号转导和转录激活因子5(STAT5)在小鼠CD4^+CD25^+调节性T细胞中的表达情况。方法:免疫磁珠法分离C57BL/6J小鼠脾脏中的CD4^+CD25^+调节性T细胞,共聚焦荧光法检测细胞内STAT5的分布并进行初步定位,进一步采用Western blot技术从蛋白水平检测细胞内STAT5的表达。结果:激光共聚焦显微镜成像显示STAT5蛋白主要存在于CD4^+CD25^+调节性T细胞胞浆中,细胞总蛋白Western blot检测显示出清楚的STAT5和磷酸化STAT5条带,分子质量分别为92 kD和94 kD。结论:STAT5在小鼠CD4^+CD25^+调节性T细胞内存在表达且主要为胞浆表达。
Objective: To investigate the expression of transcription factor signal transducers and activators of transcription (STAT) 5 in mouse CD4+ CD25+ regulatory T cells. Methods: CD4+ CD25+ regulatory T cells from the spleen of C57BL/6J mice were isolated by means of MACS microbeads, and then were evaluated with Western blot test and confocal microscopy analysis for STAT5 expression. Results: Confocal microscopy analysis showed that mouse CD4+ CD25+ regulatory T cells did express STATS protein in its cytoplasm. Through Western blotting it was revealed distinct bands of STAT5 and p-STAT5, and their molecular weight was 92 kD and 94 kD respectively. Conclusion: STAT5 protein can be markedly expressed in mouse CD4+ CD25+ regulatory T cells, and it is mainly located in the cytoplasm of these cells.
