摘要 目的 通过对ELISA方法影响因素的分析,对酶联免疫反应加速仪进行评价.方法 应用正交试验设计对ELISA反应外界影响因素(温度、振动频率、反应时间)的优化,以检测HBsAg为例来进行评价.结果 在ELISA反应的三个影响因素中,振频和反应时间对结果有显著性影响(P〈0.05),温度虽对结果有一定的影响但无显著性意义;酶联免疫反应加速仪检测HBsAg的最低检出限与常规法都为1 ng/ml,二者差异无统计学显著性意义(P>0.05).结论 酶联免疫反应加速仪能够加速反应进行,反应的灵敏度和阳性检出率符合检测要求,而且具有良好的重复性和特异度. Objective To estimated the ELISA-accelerate-equipment by analysing the effectors of ELISA. Methods Selected the best experimental condition of ELISA by analysing the effective factors by Orthogonal Experiment Design,to ap- praise the assay through detection of HBsAg. Results Among the three effect factors,vibrate frequency and reaction time had significant effect on ELISA results(P〈0. 05),while the temprature had nonsignificant effect although it had some effect (P〉0. 05);The detection limits of HbsAg by vibrate ELISA and normal method were lng/ml both. There was no significant difference between two assay(P〉0. 05). Conclusion The results showed that the ELISA-accelerate-equipment could accelerate the reaction between antigen and antibody ,and the sensitivity and detection rate of positive could fit the requirement of clinical detection, and it has good replication and specification.
