摘要
目的 探讨胆囊收缩素B受体(CCK-B)在人结肠癌组织及结肠癌旁组织中的表达情况,以筛选出有效的结肠癌生物学标志.方法 收集外科手术切除结肠癌组织标本41例和对应癌旁正常结肠组织,采用RT-PCR法检测结肠癌和癌旁组织中CCK-B受体mRNA的表达;采用免疫组化染色法检测CCK-B受体蛋白在结肠癌组织中的表达水平以及与临床分期和病理分级之间的关系.结果 经RT-PCR检测41例结肠癌组织中有36例CCK-B受体mRNA呈阳性,阳性表达率为87.8%;癌旁组织CCK-B受体mRNA有10例表达,阳性表达率为24.4%.两组间CCK-B受体mRNA阳性表达率经统计学处理,差异有统计学显著性意义(P<0.05).免疫组化检测显示41例结肠癌组织中CCK-B受体蛋白有34例呈阳性表达,阳性表达率为82.9%,癌旁组织CCK-B受体蛋白有6例表达,阳性表达率为14.6%,两组间CCK-B受体蛋白阳性表达率经统计学处理,差异有统计学显著性意义(P<0.05).结肠癌组织不同病理学分级各组间CCK-B受体阳性表达率经统计学处理,差异无统计学显著性意义(P〉0.05).结论 CCK-B受体在肿瘤的发生发展过程中起一定的作用,CCK-B受体的表达可能是结肠癌发生的早期事件,可作为结肠癌临床诊断的生物学标志,并为结肠癌的临床治疗提供了有效的靶受体.
Objective To investigate the expression of eholecystokinin receptor B type (CCK-B) in colon cancer tissues and normal tissues. Screening effective biological marker in colon cancer tissues. Methods The expression of CCK-B receptor mRNA in 41 colon cancer tissues and in tumor adjacent tissues was detected by RT-PCR and the expression of CCK-B receptor protein was detected by immunohistochemistry staining. The relationship of CCK-B receptor expression with the clinical stage and pathological grade was analysed. Results There were 36 cases CCK-B receptor mRNA expression in 41 colon cancer tissues and the pesitive rate was 87.8%. There were 10 cases CCK-B receptor mRNA expression in tumor adjacent tissues. There were 34 cases CCK-B receptor expression in 41 colon cancer tissues and the positive rate was 82. 9%. There was 6 cases CCK-B receptor expression in detected tumor adjacent tissues. Significant difference was found between colon cancer tissues and in tumor adjacent tissues (P〈0. 05). No significant difference was found among the clinical stage ahd pathological grade groups(P 〉0. 05). Conclusion CCK-B receptor play an important role in the incidence and devel- opment of colon cancer and probably would be an early event in the colonic carcinogenesis. It maybe serve as novel tumor marker in clinical diagnosis for colon cancer and offer an effective therapeutic target receptor of the disease.
