医药废水中大肠杆菌抗体制备及检测方法 被引量:1

摘要 该文研究制备了大肠杆菌全菌体抗体,经纯化后抗体蛋白含量为2.50 mg/mL,测得全菌体抗体的效价大于25 600,并使用自制的抗体,建立了测定医药废水中大肠杆菌的间接酶联免疫吸附(ELISA)方法。间接ELISA方法的最佳测定条件为:抗原最佳包被浓度为200倍,一抗和酶标二抗的稀释浓度分别为稀释12 800倍和2 000倍,以0.5%牛血清白蛋白(BSA)作为封闭液,封闭时间为2 h。在最佳测定条件下,大肠杆菌在1×10^3-1×10^9 cfu/mL表现出良好的线性关系。采用所建立的间接ELISA方法检测了实际医药废水中的大肠杆菌,检测限为10^3 cfu/mL。 In order to detect escherichia coli in pharmaceutical wastewater quickly,indirect ELISA method was established.Preparation of escherichia coli antibodies,antibody protein content after purified was 2.50 mg / mL,the titer of all bacteria antibody was more than 25 600.The optimal detection conditions are as follows: the concentration of antigen is 200 times;the diluted concentration of antibody and enzymes standard antibody are 12 800 times and 2 000 times,respectively;0.5 % BSA is the confining liquid.Under such detection condition,escherichia coli of 1×10^3-1×10^9 cfu / mL show good linear relationship.The detection limit with indirect ElISA method is 10^3 cfu/mL.
