摘要
目的:探讨CT引导下经皮穿刺组织块注射制作兔VX2肝癌模型方法,并与传统开腹组织块种植法进行比较;并用MSCT对模型进行评价。方法:中国大耳白兔16只,随机分成两组,每组8只,分别采用CT引导下经皮穿刺瘤组织块肝脏注射及开腹直视下瘤组织块肝脏接种的方式进行成瘤。肿瘤种植后第15 d,进行CT平扫及增强扫描评价肿瘤接种成功率及生长情况,并观察两组动物感染率差别。结果:肿瘤接种成功率两组均为87.5%。肿瘤最大直径穿刺组和开腹组分别为(1.16±0.23)cm和(1.28±0.27)cm,两种方法成瘤大小差异无统计学意义。肿瘤种植后发生感染率开腹种植组(感染率42.3%)明显高于经皮穿刺组(感染率0),差异有统计学意义(P<0.05)。结论:CT引导下经皮穿刺注射瘤组织块建立兔VX2肝癌模型具有方法简单、成功率高、动物损伤小等特点,可以取代传统开腹直视下肝脏接种造模方式,有利于安全、高效地提供符合实验要求的肝癌动物模型,为肝癌临床治疗和相关基础研究做出贡献。
Objective:To explore the reproduction method of rabbit VX2 liver tomor model by percutaneous injection of VX2 tumour mass suspension into rabbit liver guided by MSCT,and compare the difference of this method with conventional one via laparotomic route,as well as evaluate the models with MSCT.Methods: 16 Chinese white rabbits were randomly divided into two arms,8 for each.VX2 tumor mass suspension via percutaneous puncture-inoculation method guided by MSCT and laparotomic route method were used to reproduce rabbit VX2 liver cancer models.Plain and contrast enhancement MSCT scan were performed to observe and evaluate the success rate of the animal models and the condition of tumor growth from the fifth day after inoculation.And the difference in infectious rate between the two arms was observed and documented for further analysis.Results:The success rates of transplanted tumor were both 87.5%;The maximum diameters of the tumors in the two groups of via laparotomic route and percutaneous puncture-inoculation methods were(1.16±0.23)cm and(1.28±0.27)cm,respectively;There was no statistically significant difference between the two methods concerned about tumor size;The infectious rate in arm of via laparotomic route method(42.3%) was significantly higher than that in arm of percutaneous puncture-inoculation method(0%) post-operation(P0.05).Conclusion:MSCT-guided percutaneous puncture-inoculation of tumor mass suspension method for reproduction of rabbit VX2 liver cancer model is simple and high efficient as well as safe to model animal,compared with the conventional method via laparotomic route,and could substitute the conventional method to serve as an safe and highly efficient one in reproduction of VX2 liver cancer animal model to meet the experimental needs and facilitate the basic research works on clinical diagnosis and treatment of liver cancer in human so as to benefit human healthy.
