摘要
用Western blot法检测青蒿琥酯对白血病K562细胞的X染色体连锁凋亡抑制因子(X-linked inhibitor of apoptosis protein,XIAP)、存活素(Survivin)、细胞凋亡抑制蛋白1(cellular IAP1,cIAP-1)和细胞凋亡抑制蛋白2(cellular IAP2,cIAP-2)表达的影响,以及对K562细胞半胱氨酸天冬氨酸酶3(caspase-3)和多聚ADP-核糖聚合酶裂解片段(poly ADP-ribose polymerase cleavage,PARP cleavage)的作用.青蒿琥酯能够下调XIAP,Survivin和cIAP-2的表达,上调caspase-3活化亚基(17kD)和PARP cleavage(85kD)的表达.青蒿琥酯通过下调XIAP,Survivin和cIAP-2的表达来诱导白血病细胞凋亡,caspase-3也与诱导白血病细胞凋亡有关.
Western blot was performed to detect the influence of Artesunate on the expression of X-linked inhibitor of apoptosis protein (XIAP), survivin, cellular IAP1 (cIAP-1), and cellular IAP2 (cIAP-2), and to investigate the effect of Artesunate on caspase-3 and poly ADP-ribose polymerase (PARP) cleavage in human erythroleukemia K562 cells. Artesunate downregulated the expression of XIAP, survivin, and cIAP-2, and upregulated the expression of a 17-kD caspase-3 cleavage fragment and an 85-kD PARP cleavage fragment. By down-regulating XIAP, survivin, and cIAP-2, Artesunate induced apoptosis in K562 cells. The mechanism of this apoptosis may involve caspase 3.
