摘要
目的检测非小细胞肺癌癌组织和癌旁正常肺组织中缺氧诱导因子(HIF)-1α、P-糖蛋白(P-gp)及环氧化酶(COX)-2的表达,分析其与非小细胞肺癌生物学行为的关系。方法采用免疫组织化学染色法检测P-gp和COX-2在非小细胞肺癌癌组织的表达,用原位核酸分子杂交方法检测HIF-1α在非小细胞肺癌癌组织中的表达。结果 HIF-1α、P-gp及COX-2在肺癌组织中表达阳性率高于癌旁正常肺组织,其在非小细胞肺癌组织的表达与临床分期、肿瘤细胞分化程度、淋巴结转移、预后相关。结论 HIF-1α、P-gp、COX-2在非小细胞肺癌组织中高表达,其与非小细胞肺癌的临床特征密切相关,有可能作为肺癌不良预后的评估指标;HIF-1α、P-gp、COX-2在非小细胞肺癌组织的表达有高度相关性,为通过调节HIF-1α和COX-2的表达逆转肿瘤耐药及改善非小细胞肺癌预后的可能性提供了依据。
Objective To study the expression of HIF-1α,P-gp and COX-2 in 60 cases with non-small cell lung cancer tissues specimens and adjacent normal lung tissues specimens,and to analyze the relationship between their expression and the clinical pathological features of patients with NSCLC.Methods Immunohistochemical staining was used to detect P-gp and COX-2,in situ nucleic acid hybridization to detect HIF-1α in 60 cases of NSCLC tumor tissues and 30 adjacent normal lung tissues.Results HIF-1α,P-gp and COX-2 between lung cancer tissues and adjacent normal lung tissue were statistically significant.HIF-1α,P-gp and COX-2 expression were associated with differentiation,stage and prognosis.HIF-1α,P-gp and COX-2 in NSCLC tissues were significantly higher than that in adjacent normal lung tissue.HIF-1α,P-gp and COX-2 involved in the pathogenesis of NSCLC,differentiation,stage,prognosis.Conclusion There is close correlationship between the expression of consistency in HIF-1α,P-gp and COX-2.It implies HIF-1α and COX-2 multidrug resistance between tumor and the possibility of improving the prognosis for the HIF-1α and COX-2 inhibitors in cancer therapy provide the basis for targeting.
