真武汤对帕金森病模型大鼠学习记忆能力的影响 被引量:2

摘要 目的研究真武汤对MPTP致帕金森病(PD)模型大鼠学习记忆能力的影响。方法 SD大鼠分为假手术组、模型组、美多芭组和真武汤低、中、高剂量组,采用脑内黑质部位定向注射MPTP的方法造模,用跳台实验和Y迷宫实验评价大鼠的学习记忆能力,用高效液相色谱法分别检测模型鼠大脑皮质和海马内的多巴胺(DA)、二羟基苯乙酸(DOPAC)、5-羟色胺(5-HT)、5-吲哚乙酸(5-HIAA)和高香草酸(HVA)的含量。结果真武汤明显延长PD大鼠跳台实验的错误潜伏期,显著减少错误次数,并提高PD大鼠在Y迷宫中的自主选择正确率,且有一定的剂量依赖关系。给予真武汤后模型鼠大脑皮质内DA、DOPAC、5-HT、5-HIAA和HVA的含量明显升高,但海马内各递质含量未见显著变化。结论真武汤改善PD大鼠的学习记忆障碍作用与保护脑内多巴胺能神经系统有关。 OBJECTIVE To investigate the effect of Zhen-Wu-Tang(ZWT)on learning and memory of PD rats.METHODS SD rats were randomly divided into 6 groups:shamoperated,model,Madopar,ZWT groups of low,middle and high dosages.The PD models were induced by injecting MPTP into substantia nigra,step-down and Y-maze tests were applied to assess the capacity of learning and memory in rats;DA,DOPAC,5-HT,5-HIAA and HVA in frontal cortex and hippocampus were determined by high performance liquid chromatography(HPLC).RESULTS ZWT extended the latency and reduced the number of errors in the step-down experiment significantly,and increased the accuracy of the continuous spontaneous alternation behavior in PD rats of Y-maze.The effects mentioned above were dose-dependent.The levels of DA,DOPAC,5-HT,5-HIAA and HVA in frontal cortex of PD rats were increased after the treatment of ZWT,but these were not significantly changed in hippocampus.CONCLUSION The effect of ZWT improved learning and memory ability of PD rats associated with the protection of dopaminergic neurons in brain.
