摘要
目的探讨二氮嗪预处理对大鼠骨髓源性心肌干细胞(MCSCs)在氧化应激条件下的作用。方法利用过氧化氢溶液(H2O2)处理MCSCs造成氧化应激损伤,Annexin V/PI双标记法检测细胞凋亡的情况,采用RT-PCR和免疫印迹检测二氮嗪预处理对细胞的基因表达和Akt磷酸化水平的影响。结果经二氮嗪预处理后,在氧化应激条件下MCSCs凋亡率下降,细胞的肝细胞生长因子和碱性成纤维生长因子mRNA表达显著增加,磷酸化Akt水平明显上调。结论二氮嗪预处理可以提高大鼠MCSCs的抗氧化应激能力,其机制与提高细胞的HGF和bFGF基因表达和Akt磷酸化水平有关。
Objective To explore the effect of diazoxide(DZ) preconditioning on bone marrow-derived cardiac stem cells(MCSCs) under oxidative stress and its mechanism.Methods The oxidative stress of MCSCs was caused by H2O2.With Annexin V / PI double staining,it was detected the effect of DZ preconditioning on MCSCs under oxidative stress.The application of RT-PCR and Western blot to explore the mechanism of the effect.Results There was a significantly increase of the number of viable cells in DZ group compared with H2O2 group.HGF and bFGF expression and phosphorylated Akt of the cells was significantly raised under oxidative stress after pretreatment with DZ.Conclusion MCSCs pretreatment with DZ can reduce cell oxidative stress damage,which may be related to paracrine improvement.
