Twist蛋白上皮性钙黏蛋白mRNA及蛋白在喉鳞癌中表达的研究 被引量:4

摘要 目的观察Twist蛋白、上皮性钙黏蛋白mRNA及蛋白在喉鳞癌组织中的表达及其与临床病理特征的关系,并探讨其在喉鳞癌发生、发展中的作用及临床应用价值。方法应用原位分子杂交检测上皮性钙黏蛋白mRNA和免疫组织化学SP法检测Twist蛋白、上皮性钙黏蛋白分别在80例喉鳞癌、30例不典型增生和20例慢性炎症组织中的表达。结果①Twist蛋白和上皮性钙黏蛋白与喉鳞癌TNM分期、甲状软骨受累、病理分级、淋巴结转移和远处转移有关(P<0.05),与性别、年龄、临床分型无关(P>0.05),但上皮性钙黏蛋白mRNA的表达与病理分级无关(P<0.05)。②Twist蛋白和上皮性钙黏蛋白在喉鳞癌组织中的表达呈负相关(r=-0.317,P<0.05);上皮性钙黏蛋白mRNA和上皮性钙黏蛋白在喉鳞癌组织中的表达呈正相关(r=0.607,P<0.05)。结论①联合检测上皮性钙黏蛋白mRNA和上皮性钙黏蛋白在喉鳞癌中的表达,可以为临床早期诊断和治疗提供可靠的生物学指标。②对Twist蛋白和上皮性钙黏蛋白的深入研究有助于揭示肿瘤转移的发生机制,进而促进肿瘤的防治。 Objective To investigate the expression of Twist protein,E-cadherin mRNA and protein in laryngeal squamous cell carcinoma(LSCC) and their relation to clinicopathologic characteristics,and to explore their effects and clinical applications in the development,progress in LSCC.Methods Hybridization in situ was used to detect E-cadherin mRNA,and immunohistochemical staining SP method was used to detect the expression of Twist and E-cadherin protein in 80 cases of LSCC,30 cases of squamous displasia and 20 cases of chronic inflammation.Results ①The expression of Twist and E-cadherin was related to TNM stage,thyroid cartilage involvement,pathologic grades,and lymph node metastasis of LSCC(P0.05).②The expression of Twist was negatively correlated to the expression of E-cadherin(r=-0.317,P0.05).The expression of E-cadherin mRNA was positively correlated to the expression of E-cadherin(r=0.607,P0.05).Conclusion ①Combined detection for the expression of E-cadherin mRNA and E-cadherin in LSCC might provide reliable biological parameters for clincal early diagnosis and treatment.②Further study on Twist and E-cadherin might conduce to explain the mechanisms of neoplastic metastais and then promote the prevention and cure of neoplasms.
