摘要
采用RP-HPLC测定不同产地粉葛中葛根素和大豆苷元两种有效成分的含量。色谱柱为Kromasin C18柱(150mm×4.6mm,5μm);流动相为甲醇[B]-水[A],0—35min B相由20%升到55%;流速为0.8 mL/min;检测波长为268nm。葛根素在8.0—80.0μg/mL(r=0.9996)、大豆苷元在0.81—8.15μg/mL(r=0.9998)浓度范围内线性关系良好,葛根素的平均回收率为99.30%,RSD为0.73%,大豆苷元的平均回收率为99.54%,RSD为2.81%。RP-HPLC可以作为简单可行的粉葛药材质量评价方法。
The contents of puerarin and daidzein in different Pueraria thomsonii samples from various producing areas were determined by RP-HPLC.The separation conditions were as follows: column was Kromasin C18(150mm×4.6mm,5μm),mobile phase was methanol[B]-water[A],concentration of mobile [B] increased from 20% to 55% along with time between 0 and 35 min,flow rate was 0.8mL/min and detection wavelength was 268nm.The linear relationship of puerarin and daidzein were obtained in the range of 8.0—80.0μg/mL(r=0.9996) and 0.81—8.15μg/mL(r=0.9998),respectively.Their average added standard recovery was 99.30%(RSD=0.73%) and99.54%(RSD=2.81%),respectively.The method can be applied to quality evaluation and control of Pueraria thomsonii by RP-HPLC.
