摘要
目的:建立同时测定党参中党参炔苷及苍术内酯Ⅲ含量的HPLC分析方法。方法:采用Symmetry C18(4.6 mm×250 mm,5μm)色谱柱,流动相为乙腈(A)-水(B),梯度洗脱(0-25 min,10%A→40%A;25-35 min,40%A→100%A),流速0.8 mL·min^-1,检测波长220 nm,柱温30℃,进样量10μL。结果:党参炔苷与苍术内酯Ⅲ分离良好,进样量分别在0.076-1.52μg(r=0.9997)与0.088-1.76μg(r=0.9998)范围内线性关系良好,平均回收率分别为99.81%(RSD=1.1%)及100.1%(RSD=0.94%)。结论:该方法操作简便、准确,具有良好的重复性,可作为党参中党参炔苷与苍术内酯Ⅲ同时测定的有效方法。
Objective:To develop an RP-HPLC method for simultaneous determination of lobetyolin and atractylenolide Ⅲ in Radix Codonopsis.Methods:Symmetry C18(4.6 mm×250 mm,5 μm)was adopted,the mobile phase was acetonitrile and water with the linear gradient elution(0 min,10% acetonitrile;25 min,40% acetonitrile;35 min,100% acetonitrile)at the flow rate of 0.8 mL·min-1,the UV detector was set at 220 nm,the column temperature was 30 ℃ and the injection volume was 10 μL.Results: The resultant correlation coefficients of lobetyolin and atractylenolide Ⅲ were 0.9997 and 0.9998 with range from 0.076 to 1.52 μg,from 0.088 to 1.76 μg,respectively.The average recoveries of lobetyolin and atractylenolide Ⅲ were 99.81% with RSD of 1.1%,and 100.1% with RSD of 0.94%,respectively.Conclusion:This method can be applied to the analysis of lobetyolin and atractylenolide Ⅲ in Radix Codonopsis.The results indicated this method is simple,rapid and accurate.
