摘要
目的:观察何首乌中2,3,5,4’-四羟基二苯乙烯-2-O-β-D葡萄糖苷(二苯乙烯苷)对于大鼠肝脏酶及蛋白等生化指标的影响,以期探求何首乌"肝损伤"的物质基础。方法:大鼠分别按剂量150、300、600mg/kg灌胃给药,连续90d,恢复15天;分别于第60天、90天、105天时,全自动生化分析仪检测大鼠血清中肝酶及蛋白的含量。结果:与空白组比较,给药第60天,各剂量组球蛋白(GLB)显著升高,白球比(A/G)显著降低;给药第90天,中剂量与大剂量组的谷丙转氨酶(ALT)和谷草转氨酶(AST)显著升高,中剂量组白蛋白(ALB)显著降低;停药第15天,乳酸脱氢酶(LDH)显著降低,其他指标无统计学意义。结论:二苯乙烯苷大剂量长期使用时,会对肝脏造成一定的损伤,停药后可恢复肝功正常。
Objective : To study the effects of 2,3,5,4'- tetrahydroxystilbene - 2 - O - beta - D - glueoside ( THSG), extracted from the roots of Polygonum multiflorum Thunb. (PMT) on hepatic enzymes and serum albumin biological parameters of rats, and to further investigate its material base on protecting and injuring liver of PMT. Methods : The rats were infused with condensed THSG, different dosages (150,300,600mg/kg) once a day. It was successively administered for 90 days, and the recovery period was 15 days. The blood samples were taken on the 60d,90d, and 105d respectively for biochemical examination. Results:Compared with control group,THSG can increase GLB, and reduce A/G in 60d. The mild and large dosage of THSG can increase ALT,AST significantly ( P 〈 0.05 ) in 90d. Conclusion : Long term use of large dose THSG for intragastric administration does injure to liver, which had been relieved or disappeared during the medication stopped.
