摘要
背景:已证实姜黄有广泛的抗炎与抗氧化作用,对血管内皮有保护功能。目的:观察姜黄煎剂对内皮细胞组织损伤模型大鼠热休克蛋白27(HSP27)及肿瘤坏死因子α(TNF-α)的影响,从炎性反应的角度在分子水平探讨姜黄煎剂保护血管内皮细胞的机制。方法:模拟PCI术,采用ELISA法检测姜黄煎干预组、生理盐水干预组、假手术组与健康组大鼠血清HSP27及TNF-α的水平含量。结果与结论:结果姜黄煎干预组的HSP27与TNF-α的表达均与其他各对照组有明显差异(P﹤0.05),且HSP27在3~7天时表达高于其他对照组,TNF-α的表达各时期均低于模型组和假手术组,高于正常组。模型组3d天后组织染色光镜观察,姜黄组相同时间点损伤程度明显低于模型组。推断:大鼠损伤后炎性因子TNF-α及HSP27的表达表明姜黄煎剂作用机制可能与抑制TNF-α及上调HSP27水平表达而发挥保护血管内皮的作用。
Background:Previous studies demonstrated that cureumin has widely anti - inflammatory effect and protects vascular endothelial cell against injury. Objective : To observe the effects of curcuma decoction on heat shock protein 27 (HSP27) and tumor necrosis factor α (TNF -α) in rats with vascular endothelial injury, and to explore the molecular mechanism of protection effects from the aspect of inflammatory reaction. Methods : Wistar rats were randomly divided equally into normal control, sham operation, model and curcuma groups. The expressions of HSP27 and TNF - α were measured by ELISA assay. The morphological changes of vascular endothelial cells were observed by hematoxylin - eosin staining. Results : andConclusion : The expressions of HSP27 and TNF -α were significantly diffrient from other groups. ( P 〈 0.05 ). The expressions of HSP27 were higher than other groups at 3 and 7 days ,the expressions of TNF -α were lower than those in the model and shamp operation groups at 3,7and 14 days (P 〈0.05).
