摘要
为探讨甲基斑蟊胺对HepG2细胞凋亡的诱导作用,揭示其抗癌机制,用MTT法观察药物对细胞的生长抑制作用,用TUNEL法、流式细胞仪及光镜等技术研究癌细胞凋亡。结果表明药物对癌细胞有明显的抑制生长作用;光镜可见染色质浓缩、核碎裂等改变;流式细胞仪可检测到凋亡峰;6mg/ml药物作用5天,TUNEL法强阳性。提示甲基斑蟊胺可诱导HepG2细胞凋亡。
To demonstrate the induction of apoptosis of human HepG_2(Hepatocellular Carcinoma) cell lineN-methylcantharidinimidum in order to investigate the mechanism of its anticancer action. MTT method,TUNIflow cytometry(FCM) and light microscopy were used. HepG2 cells treated with N-ethylcantharidinimidumunderwent cell proliferation inhibition;typical nuclear condensation and fragmentation can be detected by lightmicroscopy;apoptotic peaks were also detected on FCM,and the TUNEL-positive cells can be seen after beingtreated with N-methylcantharidinimidum at 6mg/ml for 5days. N-methylcantharidinimidum can induce apoptosisof HepG2 cells in a certrain concentration.
