摘要
目的:通过黄芩干预热病征候的代谢组学研究,阐明药性与功效之间的关联性,初步揭示中药性效关系的科学内涵。方法:应用代谢组学技术,主成分分析和正交偏最小二乘-判别分析方法,运用UPLC/EST-TOF/MS检测手段,研究2,4-二硝基苯诱导的热病证候代谢特性及黄芩的干预作用。结果:在主成分得分图中造模后大鼠尿液代谢物组发生变化且被明显分为2类,这从体内微观角度表征了空白组和模型组大鼠代谢网络的异同,同时确定了38个表征热病证候发生、发展的特异性生物标志物,给予黄芩干预后大鼠尿液中的代谢物组在主成分得分图上处于模型恢复的路线上,并对7个表达上调的离子具有明显的回调作用。结论:本研究从热病证候代谢物组的整体及生物标志物的变化表征了黄芩的苦寒性效关系,同时也为中药传统药性理论的研究提供了新的思路、方法和技术手段。
Objective:To study the metabonomics of intervention of Radix Scutellariae on fever syndrome induced by 2,4-dinitrophenol(DNP),and to clarify the relevance between the drug properties and the effectiveness,and preliminarily reveal the scientific connotation of property-effect relationship of Chinese medicinal herbs.Methods:The metabonomic technique and PCA and OPLS-DA analysis methods,the test facility of ultra-performance liquid chromatography/electrospray ionization quadruple time-of-flight mass spectrometry(UPLC/ESI-Q-TOF-MS) were utilized to study the metabolic characteristics of fever syndrome iduced by DNP and intervention effect of Radix Scutellariae.Results:In score plots urinary metabolites changed significantly after modelling and divided into two categories,which displaying the similarities and differences between the metabolic networks of two groups from microscopic view in vivo.Meanwhile thirty eight specific biomarkers were determined which could represent fever syndrome in rats.Metabolites of experimental group clustered at the route of models recuperating after the intervention of Radix Scutellariae,and it indicated that Radix Scutellariae accelerated the recuperation of fever syndrome model and made them develop in the direction of health.Seven increased ions reduced significantly in high dose group,compared with model group.Conclusion:The bitter cold relationship of property-effect of Radix Scutellariae were represented by studying the changes of the whole metabolites and biomarkers of fever syndrome.Meanwhile this subject provided a new way,methods and technical means for the study of the drug property theory of traditional Chinese medicinal herbs.
