补肾中药对地塞米松诱导的骨质疏松大鼠肾组织TRPV5表达的影响 被引量:4

Effects of nourishing-kidney herbs on expression of TRPV5 in kidney of rats with osteoporosis induced by dexamethasone

摘要 目的:通过观察补肾中药对地塞米松诱导的骨质疏松大鼠肾组织TRPV5 mRNA和蛋白表达的影响,探讨糖皮质激素性骨质疏松症(GIO)的病理机制,以及补肾中药的疗效机制。方法:地塞米松肌肉注射复制GIO大鼠模型,实验设正常组、模型组、补肾组、健脾组、活血组及骨疏康组。造模及给药9周后,采用实时定量PCR法及Western Blot法测定肾组织TRPV5 mRNA和蛋白表达。结果:与正常组比较,模型组大鼠骨密度明显降低(P<0.01),肾组织TRPV5 mRNA与蛋白表达均下降(P<0.01);与模型组比较,补肾组骨密度明显升高(P<0.01),TRPV5 mRNA与蛋白表达亦明显上调(P<0.01)。结论:补肾中药通过上调肾组织TRPV5 mRNA与蛋白表达而促进肾钙重吸收,达到治疗GIO的作用。 Objective: To observe the effects of nourishing-kidney herbs on the expression of TRPV5 in kidney of rats with dexamethasone induced osteoporosis, research the mechanism of glucocorticoid-induced osteoporosis, and the therapeutic effect of nourishing-kidney herbs. Methods: By intramuscular injection of dexamethasone to replicate rat models. Rats were divided into normal group, model group, nourishing-kidney group, invigorating-spleen group, promoting blood flow group and Gushukang group. After 9 weeks, to detect the mRNA and protein expression of TRPV5 in kidney by Real-time quantitative RT- PCR and Western Blotting methods. Results: Comparing with the normal group, the Bone Mineral Density (BMD), mRNA and protein expression of TRPV5 of model group rats decreased evidently (P〈0.01). Comparing with the model group, the mRNA and protein expression of TRPV5 of nourishing-kidney group rats increased sharply (P〈0.01), and the BMD increased evidently (P〈0.01). Conclusion: Nourishing-kidney herbs can upregulate the mRNA and protein expression of TRPV5 in kidney to promote renal calcium reabsorption, so can have the treatment effect.
