艾灸预处理对心肌缺血高脂血症大鼠心肌RyR2 mRNA的影响 被引量:7

摘要 目的:研究艾灸预处理对心肌缺血再灌注损伤高脂血症大鼠心肌肌浆网兰尼碱受体蛋白2(ryanodine receptor 2,RyR2)mRNA表达的变化,并与缺血预处理的效果进行比较,探讨艾灸预处理的有效性和对心肌的保护机制。方法:将高脂血症SD大鼠随机分为假手术组、缺血预处理组、艾灸预处理组和缺血再灌注组,采用推管法复制大鼠心肌缺血再灌注损伤模型,对心肌进行荧光半定量RT-PCR检测,测定RyR2 mRNA表达水平的变化。结果:与假手术组比较,艾灸预处理组及缺血预处理组心肌肌浆网RyR2 mRNA表达均显著下降,但高于缺血再灌注组(P<0.01)。结论:艾灸预处理与缺血预处理均可减轻心肌缺血再灌注损伤诱导心肌RyR2 mRNA表达的下调,减轻心肌缺血再灌注对心肌造成的损伤,丰富了"治未病"理论。 Objective:To observe the changes of mRNA expression of ryanodine receptor 2 within sarcoplasmic reticulum,from model rats of ischemic reperfusion injury hyperlipemia by moxibustion pretreatment.And compare the results with ischemic preconditioning to discuss the efficiency of moxibustion pretreatment and the protecting mechanism of myocardium.Methods:Randomly divided hyperlipemia SD big rats in to a sham group,an ischemic preconditioning group,a moxibustion pretreating group and an ischemic reperfusion group.Establish ischemia reperfusion injury model rats by pushing tube detected myocardium with RT-PCR to determine changes of RyR2mRNA expression.Results:Compared with sham group,RyR2mRNA expression in ischemic preconditioning group and moxibustion pretreating group were both decreased significantly,while still higher than ischemic reperfusion group(P〈0.01).Conclusion:Moxibustion pretreating and ischemic preconditioning can both lower the decreasing of RyR2mRNA expression,and reduce the injury of ischemic reperfusion,and that enriches the theory of preventive treatment of disease.
