摘要
目的:通过检测大鼠肝、骨骼肌AdipoR1及AMPK mRNA表达,探讨黄芪有效部位(黄酮、多糖、皂苷)调节糖尿病模型大鼠血糖的可能机制。方法:SD大鼠随机分为正常组、模型组、黄芪组、黄酮组、多糖组、皂苷组,观测30d,检测血糖、血清胰岛素(ELISA),肝、骨骼肌AdipoR1、AMPK mRNA表达(real-time PCR)。结果:模型组血糖水平升高,血清胰岛素水平及肝、骨骼肌组织中AdipoR1、AMPK mRNA表达显著下降(P<0.01);黄芪各有效部位均可降低血糖、升高血清胰岛素水平,以黄芪组、黄酮组作用显著(P<0.05);黄芪各有效部位可上调肝和骨骼肌组织中AdipoR1、AMPK mRNA表达,其中在肝组织中以黄芪组、黄酮组升高明显,在骨骼肌组织中以皂苷组升高明显(P<0.05)。结论:黄芪有效部位可降低糖尿病模型大鼠血糖、升高血清胰岛素水平,以黄芪黄酮效果显著;其调节血糖的机制可能与肝、骨骼肌AdipoR1、AMPK mRNA表达水平变化有关。
Objective:This study was designed to discuss the possible mechanism in the procedure of Astragalus active ingredients(includes Astragalus flavonoids,polysaccharides,and astragalosides) adjusting blood glucose in diabetic rats by assaying AdipoR1,AMPK mRNA expression in liver and skeletal muscle.Methods:SD rats were randomly divided into normal group,model group,Astragalus group,Astragalus flavonoids group,polysaccharides group,and astragalosides group.After 30d,the detection of blood glucose,ELISA assay of serum insulin,adipoR1 and AMPK mRNA expression in liver and skeletal muscle should have be done.Results:In model group,blood glucose increased,serum insulin and adipoR1,AMPK mRNA expression in liver and skeletal muscle decreased significantly(P〈0.01).All of Astragalus active ingredients could reduce blood glucose and ascend serum insulin,the significant ones were anstragalus solution and Astragalus flavonoids(P〈0.05).All of Astragalus active ingredients could up-regulated adipoR1 and AMPK mRNA expression in liver and skeletal muscle,which the remarkable increases of adipoR1,AMPK mRNA expression were Astragalus solution Astragalus flavonoids in liver and astragalosides in skele muscle(P〈0.05).Conclusion:Astragalus active ingredients could regulate blood glucose and serum insulin levels in diabetic rats,the best one was Astragalus flavonoids.Its mechanism may be related to the changes of adipoR1,AMPK mRNA expression in liver,skeletal muscle.
