摘要
目的:探讨补肾抗衰片对家兔动脉粥样硬化病变后海马组织氧化应激的影响。方法:将56只日本大耳白兔随机分为正常组8只,实验组48只,正常组给予普通饲料,实验组建立动脉粥样硬化模型,在第8周时,实验组随机分为模型组、补肾抗衰片干预组、辛伐他汀干预组。各组分别于给药16周后处死,无菌条件下取脑,用Q-PCR法检海马HO-1mRNA的表达,ELISA法检测海马组织SOD、GSH-Px活性和MDA含量以及血清中SOD、MDA的水平。结果:家兔海马HO-1 mRNA基因在各组均有表达,与正常组相比,模型组海马HO-1 mRNA表达差异无统计学意义,但模型组经辛伐他汀和补肾抗衰片干预后,海马HO-1 mRNA表达均升高,差异具有显著的统计学意义(P<0.05)。ELISA法显示海马组织SOD、MDA、GSH-Px以及血清SOD、MDA的含量各组比较均有统计学意义(P<0.05或P<0.01)。结论:中药复方制剂补肾抗衰片在动脉粥样硬化所致脑损伤中具有重要的抗氧化作用,其保护机制可能是通过调控HO-1mRNA基因的表达以及影响SOD、GSH-Px酶的活性,同时清除脂质氧化终产物MDA而实现的。
Objective:To investigate the effect of Bushen Kangshuai Tablets in treating atherosclerotic lesion and the mechanism of oxidative stress in rabbits hippocampus.Methods:The 56 Japanese white rabbits were divided into normal group 8,and the experimental group 48 randomly,the normal group were given normal diet,the experimental group a reference to the establishment of experimental atherosclerosis model of oxidative stress in the first 8 weeks.Experimental group randomly divided into model group,Bushen Kangshuai group and simvastatin group.Each group were sacrificed after drug administration for 16 weeks after the brain was removed under sterile conditions,using Real Time Quantitative PCR check the expression of HO-1mRNA of hippocampus,SOD,GSH-Px and MDA concentrations in serum were detected by Enzyme-linked immunosorbent assay(ELISA).Results:HO-1 mRNA gene expressed in rabbit hippocampus in each group,there was no significant difference in the expression of HO-1 mRNA gene between the normal and the model group,but after treated with Bushen Kangshuai and simvastatin tablets,the mRNA level of HO-1 increased significantly(P〈0.05);SOD,MDA and GSH-Px were significantly higher than the normal group(P〈0.05 or P〈0.01).Conclusion:Chinese herbal compound Bushen Kangshuai Tablets in atherosclerosis induced brain injury had an important antioxidant role,the way may be through regulatied the HO-1mRNA gene expression,or may be stimulated the up regulation of SOD,GSH-Px and scavenged the MDA.
