优化活血利水方对大鼠自体髓核移植后移植髓核MMP-3、TIMP-1表达的影响 被引量:4

摘要 目的:建立自体髓核移植致大鼠非压迫腰椎间盘髓核突出神经根性疼痛模型,探讨优化活血利水方对移植髓核MMP-3、TIMP-1表达的影响。方法:40只SD大鼠随机分为对照组、模型组、中药组、阿斯匹林组,建立大鼠自体髓核移植致腰5神经根性痛模型,造模后1周起各组分别予生理盐水1.82mL、生理盐水1.82mL、中药汤剂1.82mL、阿斯匹林1.82mL(0.1g.kg-1.d-1),每日1次,连续用药4周。第31天取移植髓核检测MMP-3、TIMP-1mRNA及蛋白的表达。结果:正常髓核中MMP-3、TIMP-1mRNA及蛋白表达量很少,各实验组移植髓核中MMP-3、TIMP-1mRNA及蛋白表达量与对照组比较均显著增多(P<0.0l)。与模型组比较,中药组、西药组MMP-3mRNA及蛋白表达量显著减少(P<0.01),TIMP-1mRNA及蛋白相对表达量显著增多(P<0.01)。中、西药组比较,两组间存在显著性差异,中药组MMP-3mRNA及蛋白显著减少,TIMP-1mRNA及蛋白显著增多。结论:优化活血利水方可以显著下调移植髓核MMP-3mRNA及其蛋白表达,上调TIMP-1mRNA及其蛋白表达,减轻椎间盘的退变,促进椎间盘的修复。 Objective: To design Non - compressive model study of experimental lumbar nucleus pulposus prolapse breeding nerve root pain in rats by Transplantation of Autologous Nucleus Pulposus, to investigate the effects induced by Huoxue Lishui Fang on the expression level of MMP -3 and TINP - 1 of transplanted nucleus pulposus. Methods :40 SD (Sprague - dawley) rats were chosed. The rats were randomly divided into the control group, model group, the Chinese Medicines Group and the aspirin group. To design L5 nerve root pain model induced by Transplantation of Autologous Nu- cleus Pulposus in Rats. A week later, 1.82mL NS, 1.82mL NS, 1.82mL Decoction of Chinese medicine and 1.82mL as- pirin (0. 1g·kg-1·d -1) were given separately once a day. Medication for 4 weeks in a row. A month later,the trans- planted nucleus were taken out for testing the expression of MMP -3,TIMP - 1 mRNA and protein. Results:In normal nu- cleus pulposus , there is a small amount of MMP- 3 ,TIMP-1 mRNA and protein. Compared with the control group,the amount of MMP- 3 ,TIMP- lmRNA and protein in each experimental group increases notably (P 〈 0.01 )o Compared with model group, MMP - 3mRNA and relative protein in Chinese medicine group and western medicine group decreases. However,TIMP- 1 mRNA and prtein increases remarkably (P 〈 0.01 ). There is clear difference between Chinese medi- cine group and western medicine group. In Chinese medicine group, MMP- 3mRNA and prtein decreases remarkably,but 'lIMP- lmRNA and protein increases notably. Conclusion: Optimized Huoxue Lishtti Fang significantly downregulated the expression of MMP - 3 Protein and mRNA and up - regulated the expression of TIMP - 1 Portein and mRNA in transplanted nucleus pulposus, relieving lumbar intervertebral disc degeneration, promoting repair of lumbar intervertebral disc.
