温针灸对兔膝骨关节炎模型基质金属蛋白酶及组织形态学影响的实验研究 被引量:43

摘要 目的:观察温针灸对膝骨关节炎模型血清基质金属蛋白酶及软骨组织形态学的影响,探讨温针灸治疗膝骨关节炎在软骨代谢方面的作用机制。方法:将新西兰兔随机均分为空白组、模型组、温针灸组。采用膝关节前交叉韧带切断术制备膝骨关节炎模型,造模1周后温针灸患侧膝关节"犊鼻"、"后三里"穴2个疗程,采用ELISA法测定各组血清中MMP-1、MMP-3、TIMP-1的浓度,股骨内髁行HE染色。结果:模型组MMP-1浓度、MMP-1/TIMP-1比值均高于空白组(P﹤0.05),MMP-3浓度低于空白组(P﹤0.05),TIMP-1变化不明显(P﹥0.05),HE染色示明显的病理损伤;与模型组比较温针灸降低了MMP-1的浓度与MMP-1/TIMP-1之比值,升高MMP-3的浓度,差异有统计学意义(P﹤0.05);且改善了软骨病理损伤。结论:温针灸能够提高膝骨关节炎软骨的自身修复能力,其软骨保护作用可能是通过调节异常的MMP-1、MMP-3与MMP-1/TIMP-1比值而实现的。 Objective :To investigate the influence of warm acupuncture- moxihustion on matrix metalloproteinase and histomorphology of the rabbit experimental Knee Osteoarthritis Model, and explore its chondroprotective mechanism. Meth- ods : New Zealand rabbits were equally randomized into control, model and warm acupuncture - moxibustion groups. KOA model was duplicated by anterior cruciate ligament transaction (ACLT). After KOA model was established one week, aeupolnts of"Dubi" (ST35)," Housardi" (ST36) were used for treatment with warm acupuncture- moxlhustion, two courses totally. The concentration of MMP - 1 ,MMP -3 and TIMP - 1 in serum were detected by enzyme linked immu- nosorbent assay( ELISA). The histological changes of the medial femoral condyle was observed by haematoxylin - eosin staining. Result: In comparison with control group, the concentration of MMP - 1 and rate of MMP - 1/TIMP - 1 in- creased significantly in model group( P 〈 0.05 ) , while MMP- 3 reduced significantly( P 〈 0.05 ), also cartilage degrada- tion was more severe; compared with mode/ group, MMP - 1 and MMP - 1/TIMP - 1 ratio reduced significantly(P 〈 0; 05 ), MMP - 3 increased significantly( P 〈 0.05 ) in warm acupuncture - moxibustion groups, also cartilage degradation was not severe. Conclusion : Warm acupuncture - moxibustion can improve the capability of self - repairing of cartilage, it ehondroproteetive effect achieved by regulating the abnormal MMP- 1 ,MMP- 3 and MMP- 1/TIMP -1 ratio.
