摘要
目的:探讨复方丹参片对急性脑缺血大鼠血管新生因子影响。方法:采用栓线法制备大鼠脑缺血模型。实验分为正常组、假手术组、模型组术、血塞通片组、复方丹参片组五组,术后后正常组、假手术组、模型组术蒸馏水灌胃,血塞通片组予于血塞通片灌胃,复方丹参片组予于复方丹参片灌胃。分别在24h,3天,7天等时间点对以上5组进行神经功能缺损评分,然后断头取脑,TTC染色观察脑梗死面积,免疫组织化学方法观察梗死周围区脑组织VEGF和bFGF蛋白的表达。结果:复方丹参片可明显改善急性脑缺血大鼠的神经功能,缩小梗死面积,显著增加大鼠脑内VEGF和bFGF蛋白的表达,促进急性脑缺血大鼠脑缺血组织侧枝循环的建立。结论:复方丹参片增强急性脑缺血大鼠梗死灶周围血管生长因子的表达,促进了梗死灶周围的血管新生。
Objective : To explore the Efect of Endothelial Growth Factor of Salvia on rats with cerebral ischemia. Methods:Cerebral ischemia model after middle cerebral artery occlusion (MCAO)in rats was induced by the method of nylon monofilament via external carotid artery . artery. Rats were divided randomly into normol group, ischemia model group , sham operation group, ischemia model with Xuesaitong tablet treatmen group ,ischemia model with Compound salvia tablet treatmen group. Normol group, isehemia model group , sham operation group were treated with distilled water; isehemia model with Xuesaitong tablet treatmen group treated with Xuesaitong tablet, ischemia model with Compound salvia tablet treatmen group treated with Compound salvia tablet. Indication of rats based on observation after twenty - four hours, three days, seven days. The area of cerebral and neurological deficit were determined by TTC staining and Longa S score. VEGF and bFGF protein were detected by immunohistochemistry and in situ hybridization. Results:Compound salvia tablet has protective effects for cerebral ischemia shrink the area of cerebral through upregulating the expression of VEGF and bF- GF. The mechanism is may be it had promote collateral circulation in ischemic tissue of acute cerebral ischemia. Conclu- sion : Compound salvia tablet can increase the expression of Endothelial Growth Factor and promote the angiogenesis in rats with cerebral ischemia.
