冰糖草组织培养快速繁殖研究

摘要 【目的】对冰糖草进行组织培养及快速繁殖研究,建立冰糖草的无性繁殖体系。【方法】以冰糖草叶片、茎段侧芽、幼嫩顶芽、未开裂果实为外植体材料,考察各种外植体的最佳灭菌时间,选取最佳外植体,诱导丛生芽,考察蔗糖浓度、天然添加物、生长调节剂对冰糖草丛生芽增殖的影响,生根培养,炼苗移栽。【结果】以果实为外植体诱导丛生芽较适宜,最佳灭菌方法为体积分数75%乙醇浸泡30 s,2 g/L升汞溶液灭菌20 min。丛生芽诱导培养基以MS+2.0 mg/L 6-BA+0.1mg/L NAA为佳。丛生芽增殖的最佳培养基为MS+1.0 mg/L IBA+2.0 mg/L 6-BA+0.3 mg/L NAA,增殖率达488.68%。生根培养基为1/2 MS,生根率达100%。移栽基质为营养土,移栽成活率达80%。【结论】利用植物组织培养技术能够进行冰糖草丛生芽诱导和增殖实验,建立快速繁殖体系。 Objective To probe an efficient way for tissue culture and rapid propagation of the Scoparia dulcis L.and to establish the vegetative propagation system.Methods The leaves,lateral buds,young terminal buds,and unbroken fruits of Scoparia dulcis L.were taken as explants.With the optimal sterilization time as the observation index,the optimal explants were screened.Then the optimal explants were used for inducing cluster buds.The effect of sucrose concentration,natural additives,and growth regulator on the proliferation of cluster buds was investigated.Results The unbroken fruits of Scoparia dulcis L.could be used for the optimal explants.The optimized sterilization conditions were soaking with 75% ethanol for 30 s and sterilization with HgCl2 2 g/L for 20 min.The medium of MS+6-BA 2.0 mg/L+NAA 0.1 mg/L was suitable for shoot induction.The medium of MS + IBA 1.0 mg/L + 6-BA 2.0 mg/L + NAA 0.3 mg/L was optimal for the proliferation of cluster buds,the proliferation rate being 488.68%.The medium of 1/2 MS was suitable for inducing roots and the rooting rate was up to 100%.With the nourishment soil as the transplantation medium,the survival rate for plantlets transplantation was up to 80%.Conclusion Cluster buds induction and proliferation can be achieved in Scoparia dulcis L.through plant tissue culture,and rapid propagation system has been established.
