摘要
以鸡血清为实验材料提取农药残留快速检测酶制剂胆碱酯酶,确定了鸡血清胆碱酯酶的最佳提取和纯化工艺,即对粗酶液进行盐析、透析、吸附层析和葡聚糖凝胶柱层析等实验方法。结果表明:鸡血清胆碱酯酶经过硫酸铵盐析,透析、吸附层析和Sephadex G-100柱层析处理后,酶活力分别提高了1.548、.21、9.03、13.00倍,冷冻干燥后酶活力提高了13.58倍,比酶活力由粗酶液中的0.538 OD值/mg提高到层析后的7.304 OD值/mg。
Chicken serum was selected as experimental material.The possibility of the ammonium sulfate saturation on the effect of purification were explored.The best extraction and purification of chicken blood.The results show that： the cholinesterase enzyme activity after ammonium sulfate has increased 1.54 times,dialysis has increased 8.21 times,after adsorption chromatography has increased 9.03 times,after Sephadex G-100 column chromatography has increased 13.00 times,after freeze and cry has increased 13.58 times.Specific activity has increased to 7.304 ODvalue/mg from the crude enzyme liquid of 0.538 ODvalue/mg
