摘要 以鸡血清为实验材料提取农药残留快速检测酶制剂胆碱酯酶,确定了鸡血清胆碱酯酶的最佳提取和纯化工艺,即对粗酶液进行盐析、透析、吸附层析和葡聚糖凝胶柱层析等实验方法。结果表明:鸡血清胆碱酯酶经过硫酸铵盐析,透析、吸附层析和Sephadex G-100柱层析处理后,酶活力分别提高了1.548、.21、9.03、13.00倍,冷冻干燥后酶活力提高了13.58倍,比酶活力由粗酶液中的0.538 OD值/mg提高到层析后的7.304 OD值/mg。 Chicken serum was selected as experimental material.The possibility of the ammonium sulfate saturation on the effect of purification were explored.The best extraction and purification of chicken blood.The results show that: the cholinesterase enzyme activity after ammonium sulfate has increased 1.54 times,dialysis has increased 8.21 times,after adsorption chromatography has increased 9.03 times,after Sephadex G-100 column chromatography has increased 13.00 times,after freeze and cry has increased 13.58 times.Specific activity has increased to 7.304 ODvalue/mg from the crude enzyme liquid of 0.538 ODvalue/mg
