摘要
目的:了解T细胞受体信号传导分子CD3γδεζ基因在T和B细胞白血病病人中的表达特点。方法:采用SYBR GreenⅠ荧光定量PCR和相对定量分析法检测32例淋巴细胞肿瘤病人:急性B淋巴细胞白血病(B-ALL)12例、慢性B淋巴细胞白血病(B-CLL)9例、急性T淋巴细胞白血病(T-ALL)11例及10例健康人外周血单个核细胞中CD3各亚单位表达情况,以β2微球蛋白基因(β2M)作为内参,采用公式2-△Ct×100%计算CD3分子γ、δ、ε和ζ链表达水平。结果:B-ALL病人的CD3分子γ、δ和ε链表达中位数水平分别为4.29%,0.67%,17.8%,与健康对照组相比,无显著性差异(P>0.05);而CD3γ链表达水平为0.99%,与健康对照组相比,显著降低(P=0.02)。B-CLL病人的CD3δ、ε基因的表达分别为0.52%,6.72%,与健康对照组相比,无显著性差异(P>0.05);而CD3γ、ζ基因(2.72%,0.77%)则比健康人明显下降(P<0.05)。在T-ALL病人中,CD3分子γ、δ和ε的表达(32.99%,12.9%,61.56%)与健康对照组相比,均明显上升(P<0.05),而CD3ζ的表达量为4.17%,与健康人无显著性差异(P=0.44)。B-ALL、B-CLL病人和健康对照组的CD3分子各链的表达模式依次为CD3ε,CD3γ,CD3ζ,CD3δ,而T-ALL病人的表达模式顺序为CD3ε,CD3γ,CD3δ,CD3ζ。结论:在B细胞白血病中CD3各基因的表达水平多为降低,其变化可能与机体T细胞免疫缺陷相关。而在T细胞白血病中CD3各基因表达水平增加可能是白血病性T细胞一个特点。
Aim:In order to study the feature of T cell receptor signal transduction factor CD3 genes in patients with lymphoid malignancies.Methods: Real-time PCR with SYBR GreenⅠtechnique was used for determination of the expression levels of all 4 CD3 genes in peripheral blood mononuclear cells of 10 healthy individuals and 32 patients with lymphoid malignancies: 12 cases of including of B-cell acute lymphoblastic leukemia(B-ALL),9 cases B-cell chronic lymphoblastic leukemia(B-CLL),11 cases of T-cell acute lymphoblastic leukemia(T-ALL).β2-microglobulin gene(β2M) was obtained as an endogenous reference.Relative changes in CD3 genes expression levels were used by the 2-△Ct×100% method.Results: The median expression levels of CD3γ,δ,ε genes in B-ALL patients at 4.29%,0.67%,17.8% respectively were no significantly different from the healthy individuals(P〉0.05).But the CD3ζ gene in B-ALL(0.99%) was lower than the healthy individuals(P=0.02).In the B-CLL group,the expression levels of CD3δ,ε genes(0.52%,6.72%) were not significantly different from healthy individuals(P〉0.05),while CD3γ,ζ(2.72%,0.77%)were lower than those of the healthy ones(P〈0.05).The expression levels of CD3γ,δ,ε genes in the T-ALL group(32.99%,12.9%,61.56%) were higher than the healthy control(P〈0.05),in contrary,the expression level of CD3ζ gene had no significantly difference from healthy individuals(P=0.44).The expression pattern of CD3ε,CD3γ,CD3ζ,CD3δ was found in healthy controls as well as in B-cell leukemias,while it was changed for CD3ε,CD3γ,CD3δ,CD3ζ in T-ALL.Conclusion:Lower expression level in most CD3 genes was found in B-cell leukemias,it might be related to the T-cell immunodeficiency.And higher expression of CD3 genes might be one of the features of T-ALL.
