人CSNK1A1基因两种转录变异体CK1αLS与CK1αS在细胞株中的表达

摘要 目的:分析CSNK1A1基因两种转录变异体CK1αLS和CK1αS在人正常细胞株和人肿瘤细胞株中的表达及在肿瘤发生发展中的作用。方法:RT-PCR法半定量检测不同细胞株中CSNK1A1基因的CK1αLS与CK1αS的mRNA表达水平。结果:36个细胞株中有32个细胞株存在CK1αLS与CK1αS的表达差异。在胚肾细胞293A、2株肺纤维细胞(MRC-5与HLF)、2株肺腺癌细胞(A549与SPC-A1)、巨细胞肺癌细胞HLAmp、小细胞肺癌细胞H446、鼻咽高分化鳞癌细胞CNE1、胃癌细胞SGC7901、骨肉瘤细胞MG-63以及3株白血病细胞(HL60、MOLT-4与Daudi)中,CK1αLS比CK1αS mRNA表达量少(P<0.05);而在肺泡细胞癌细胞H1650、肺鳞癌细胞L78、大细胞肺癌细胞H460、鼻咽低分化鳞癌细胞CNE2、3株乳腺癌细胞(Bcap-37、MDA-MB-435S与MCF-7)、2株胰腺癌细胞(BxPC-3与Panc-1)、肝细胞L02、肝癌细胞HepG2、胃上皮细胞GES-1、胃癌细胞MGC-803、结肠腺癌细胞SW620、前列腺癌细胞DU145、宫颈癌细胞Hela、卵巢癌细胞HO8910、纤维肉瘤细胞HT1080、神经胶质瘤细胞U251、脐静脉内皮细胞ECV304,CK1αLS比CK1αS mRNA表达量多(P<0.05)。胰腺癌细胞Panc-1高表达CK1αLS,低表达CK1αS;胃癌细胞SGC7901低表达CK1αLS,高表达CK1αS;结肠腺癌细胞SW620 CK1αLS与CK1αS两者表达较低;肺泡细胞癌细胞H1650 CK1αLS与CK1αS两者表达较高。结论:CK1αLS和CK1αS的mRNA表达存在差异,可能与肿瘤细胞的来源和分化程度有关。 Aim:To detect the functions of casein kinase 1alpha 1(CSNK1A1 or CK1α)gene on carcinogenesis and development by studying the expression of two transcriptional splice variants CK1αLS and CK1αS of CK1α gene in normal cell lines and malignant cell lines.Methods: The mRNA expression levels of CK1αLS and CK1αS of CSNK1A1 gene were examined by RT-PCR in thirty-six cell lines.Results: There were significant differences in mRNA expression level between CK1αLS and CK1αS in thirty-two cell lines.In the following cell lines,including human kidney embryonic 293A,two lung fibroblasts HLF and MRC-5,two lung adenocarcinoma A549 and SPC-A1,human large carcinoma HLAmp,human small cell lung cancer H446,human well-differentiated nasopharyngeal carcinoma CNE1,human gastric cancer SGC7901,human osteosarcoma MG-63 and four leukemic HL60,K562,MOLT-4 and Daudi,CK1αLS mRNA levels were lower than CK1αS′s(P〈0.05).But in other cell lines,including alveolar cancer H1650,lung squamous carcinoma L78,large cell lung carcinoma H460,poorly-differentiated nasopharyngeal epithelium carcinoma CNE2,three breast cancer Bcap-37,MCF-7 and MDA-MB-435S,two pancreatic cancer BxPC-3 and Panc-1,liver L02,hepatoblastoma-derived HepG2,gastric mucosal epithelial GES-1,gastric cancer MGC-803,colonic carcinoma SW620,prostate cancer DU145,cervical carcinoma Hela,ovarian cancer HO8910,fibrosarcoma HT1080,glioblastoma U251 and umbilical vein endothelial cell ECV304,CK1αLS mRNA levels were higher than CK1αS′s(P〈0.05).Among thirty-two cell lines,Pan-1 showed hightest level of CK1αLS mRNA and lowest level of CK1αS′s.On the other hand,CK1αLS mRNA expression was the lowest and CK1αS′s was the hightest in SGC7901.High expression of both CK1αLS and CK1αS was found in H1650,and the low was found in SW620.Conclusion:The differential expression between two transcriptional splice variant CK1αLS and CK1αS may be related to cell origin and cell differentiation in thirty-two cell lines.
