人脐血间充质干细胞移植治疗大鼠急性肝损伤 被引量:5

摘要 目的:探讨人脐血间充质干细胞对急性肝损伤大鼠模型的治疗作用。方法:体外培养人脐血间充质干细胞。60只SD大鼠随机分为3组,各20只:空白对照组、模型对照组、治疗+模型组。模型对照组大鼠采用体积分数50%四氯化碳橄榄油溶液腹腔注射造成急性肝损伤疾病模型,测定ALT/AST水平、肝脏切片HE染色评估造模效果。治疗+模型组大鼠进行开腹手术,肝脏局部多点注射人类脐血间充质干细胞悬液(3×106~5×106/只),24h后采用体积分数50%四氯化碳橄榄油溶液腹腔注射诱导急性肝损伤疾病模型。分析移植后24h实验动物存活率、肝功能改善情况及移植细胞的肝内分化情况。结果:体外培养的人脐血间充质干细胞符合间充质干细胞的一般生物学特性。模型组与治疗+模型组实验动物存活率有统计学差异(P〈0.05)。两组实验动物肝功能(AST/ALT水平)间差异有统计学意义(P〈0.05)。移植细胞在受试动物肝脏内呈人类肝样细胞方向分化趋势。结论:人间充质干细胞移植可提高急性肝损伤大鼠模型的存活率。 Aim:To explore treatment effect of human umbilical cord blood mesenchymal stem cells transplanted to a rat model of acute liver injury.Methods: Human umbilical cord blood mesenchymal stem cells were cultured in vitro.60 SD rats were randomly divided into 3 groups averagely: control group、model group,and treatment +model group.Model group rats were injected intraperitoneally with 50% olive oil solution of carbon tetrachloride to induce acute liver injury,To evaluate modeling results,ALT / AST levels in serum and liver biopsy were analyzed post-model.Rats of treatment +model group were treated with human umbilical cord blood mesenchymal stem cell suspension(3×106-5×106 / only) through local multi-point injection into the liver,then the rats were induced into acute liver injury disease models with 50% olive oil solution of carbon tetrachloride 24h post-transplantation.Survival of experimental animals and liver function improvement were analyzed post-transplantation,so does differentiation of transplanted cells in the liver.Results: Human umbilical cord blood cells cultured in vitro accord with the general biological properties of mesenchymal stem cells.There was statistically significant difference between model group and treatment +model group(P〈0.05).There were statistically significant difference about AST/ALT levels between two groups as well(P〈0.05).Transplanted cells in animal liver had a differentiation tendency to human liver-like cells.Conclusion:Transplantation of human umbilical cord blood mesenchymal stem cells could improve the survival rate of the model of acute liver injury.
