摘要
目的建立同时测定清浊祛毒丸中咖啡酸、虎杖苷和丹皮酚含量的高效液相色谱法。方法采用AgilentEclipse XDB-C18色谱柱(250mm×4.6 mm,5μm),用甲醇-0.1%磷酸流动相进行梯度洗脱,流速1.0 mL/min,检测波长314 nm,进样量5μL。结果咖啡酸、虎杖苷和丹皮酚质量浓度线性范围分别是4.8~96.0μg/mL(r=0.9995,n=5),4.1~82.0μg/mL(r=1.0,n=5)和12.0~240.0μg/mL(r=0.9999,n=5),平均加样回收率分别为101.10%(RSD=1.6%,n=9),101.62%(RSD=0.8%,n=9),102.71%(RSD=1.2%,n=9)。结论该法方便、快捷、准确、灵敏、重现性好,能更全面地控制清浊祛毒丸的质量。
Objective To establish a HPLC method for simultaneous determination of caffeic acid,polydatin and paeonol in Qingzhuoqudu Pills.Methods The Agilent Eclipse XDB-C18 column(250 mm×4.6 mm,5 μm) was used with the mobile phase of methanol-0.1%phosphoric acid.The flow rate was 1.0 mL/min and the UV detection wavelength was set at 314 nm.The sample size was 5 μL.Results The linear ranges of caffeic acid,polydatin and paeonol were 4.8-96.0 μg/mL(r=0.999 5,n=5),4.1-82.0 μg/mL(r=1.0,n=5) and 12.0-240.0 μg/mL(r=0.999 9,n=5),respectively.The average recovery rates were 101.62%(RSD=0.8%,n=9) for polydatin,101.10%(RSD=1.6%,n=9) for caffeic acid and 102.71%(RSD=1.2%,n=9) for paeonol respectively.Conclusion The method is simple,rapid,accurate and sensitive with good reproducibility,which is suitable for fully controlling the quality of Qingzuoqudu Pills.
