摘要
目的建立针对TWIST1基因启动子甲基化状态的荧光定量检测方法,进而评估其在膀胱癌临床研究的应用前景。方法应用实时荧光定量PCR方法检测南通地区50例确诊膀胱癌患者和25例非膀胱癌的健康对照者的尿液标本中TWIST1基因启动子的甲基化和非甲基化含量,对其甲基化率进行对比。结果正常人的TWIST1启动子的甲基化率为8.1%~17.3%,平均值为12.3%,膀胱癌患者的甲基化率为25.2%~48.3%,平均值为38.2%,膀胱癌患者的TWIST1基因启动子甲基化率有明显的增高(P<0.05)。结论 TWIST1基因启动子区在膀胱癌中异常甲基化,能够作为有效检测膀胱癌的基因。
Objective To establish the quantitative detection methods for TWIST1 gene methylation status in order to assess its clinical research in bladder cancer application.Methods Methylation status and unmethylation status of TWIST1 promoter were detected by realtime quantitative PCR in 50 cases of bladder cancer patients in Nantong and 25 cases of non-bladder cancer urine samples,the frequency of promoter methylation of TWIST1 gene was compared.Results The frequency of promoter methylation of TWIST1 gene in normal people was 8.1% ~ 17.3%,the average value was 12.3%,the frequency of methylation in bladder cancer patients was 25.2% ~ 48.3%,the average value was 38.2%,the frequency of methylation of TWIST1 gene in bladder cancer patients was significantly higher(P〈0.05).Conclusion Promoter methylation of TWIST1 gene in breast cancer patients is markedly abnormal,so TWIST1 can be used as an effective detection genes of bladder cancer.
