摘要
目的建立高脂血症合并血管钙化大鼠实验模型。方法在灌胃38 d高脂乳剂的基础上连续3 d给予大剂量维生素D3注射液(1×106U.kg-1)灌服,继续高脂乳剂灌胃28 d处理大鼠,观察血脂、钙、磷以及胸主动脉的形态学变化。结果在高脂乳剂-大剂量维生素D3-高脂乳剂相继灌胃处理后,大鼠血清总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-c)均明显升高(P<0.01),高密度脂蛋白胆固醇(HDL-c)无明显变化,但HDL-c/TC显著降低(P<0.01);血钙明显上升(P<0.05),主动脉形成较典型的粥样硬化斑块。结论采用高脂乳剂-大剂量维生素D3-高脂乳剂相结合的方法可以成功建立高脂血症合并血管钙化大鼠模型。
Objective To estabilsh a model of hyperlipidmia with calcification in rats.Methods The rats were fed with high-cholesterol(HC) diet for 38 d,plus vitamin D3(VD,1×105 U·kg-1) injection gavaged for 3 and then were given HC for 28 d.At endpoint,the blood lipid,the content of calcium and phosphorus were measured and morphology change of thoracic aorta were determined.Results The serum total cholesterol(TC),low-density lipoprotein cholesterol(LDL-c) were significantly increased(P0.01),high density lipoprotein cholesterol(HDL-C) did not change significantly.And HDL-c/TC were decreased(P0.01) and the content of blood calcium were increased(P0.05) in HC+VD group when compared with those in control group.The atherosclerotic plaque were presented in the rats treated with HC+VD.Conclusion The rat modle of hyperlipidmia with calcification can be established by HC+VD.
